Acupuncture experimental research results are precious resources of traditional acupuncture and moxibustion.It's not only in the guidance of acupuncture treatment of disease,but also the future of acupuncture innovation research valuable resources.However,researchers find that how to get the most useful information from those resources for their own research is a problem.The main research object of experimental information research is to report the results of the literature,but these documents are scattered in the major databases.These documents have not been systematically collated and used so far.This study took the experimental study on acupuncture experiment as a research object,and does some tentative exploratory studies of the implicit information in the literature by data mining and computational methods of complex networks.Expect to provide researchers with some new ideas and methods by this study,and improve the utilization of acupuncture experimental literature information.1.ObjectWith the continuous progress of modern science and technology,acupuncture animal experimental research also gets a very well development,and produces a large amount of literature resources.These resources still have not been used very well.So,the main purpose of this study is to find approaches to make effective use of these resources,and provide a reference for the relevant neighborhood researchers.2.Method1)literature retrievalBy collecting the acupuncture animal experimental literature scattered in the CNKI,WANFANG DATA and CBM three major databases,screening review these documents one by one,5615 target literatures were finally identified as the research object.2)data collectionAccording to the contents of the literature,to set the literature data collection information table and constitute the database background table structure.To develop a data acquisition platform on the basis of information tables and enter the original data.A total of 9 data tables were presented,including the literature table,the disease model table,the acupuncture treatment table,the electro-acupuncture treatment table,the experimental effect table,the target table,the access table,the literature external feature evaluation table,the experimental literature internal characteristic evaluation table.3)data specificationThe original data are normalized and standardized processing,according to different data reference standards,include MeSH2014 Chinese topic word list,Chinese medicine topic list 2015 edition,NCBI-Gene and Protein and KEGG database.4)bibliometrics analysis of the literatureStudy the present situation of the experimental literature on acupuncture animal by bibliometrics analysis,including age distribution of years,distribution of journals,core institutions and fund support.5)network analysis of acupoint effectFirstly,obtain the data of functional score,microstructure and tissue level of acupuncture and experimental effect.The relationship between acupoints and the three-level effect index was briefly presented by using co-word analysis method.Secondly,obtain the molecular level data in the experimental effect,and then introduce the complex network analysis method and algorithm to explore the possible biological mechanism of the acupuncture effect.Thirdly,obtain the data of the disease model and apply frequency analysis to study disease category of acupuncture experiment and study the acupuncture effects in different periods of cerebral ischemia.3.ResultsThe results and conclusions are obtained by studying the above content.The details are as follows.1)The trend of simple acupuncture experiment in the past 60 years showed that the paper grows slowly before 1995 and the amount of documents was suddenly increased after 1995.In the last ten years,the amount of documents accounted for 66.8%and the growth has been steady.These papers are mainly published in acupuncture and moxibustion magazine,Journal of Traditional Chinese Medicine and Chinese medicine academic journals.A few were published in other modern medical journals and non-academic journals.The core journals are "Acupuncture Research","Chinese Acupuncture and Moxibustion","Shanghai Acupuncture and Moxibustion","Clinical Journal of Acupuncture and Moxibustion","Chinese Clinical Rehabilitation","Liaoning Traditional Chinese Medicine","Chinese Medicine","Chinese Journal of Traditional Chinese Medicine".Funds to support acupuncture experiments are mainly supported by the National Natural Science Foundation of China,the National 973 Program funded projects,the State Administration of Traditional Chinese Medicine funded projects,etc.Among them,the number of state-funded projects is the largest,accounting for 47%of the total funding and reflecting the country's attention and support for research in this field.The author of these studies from 507 research institutions,the core research institutions in addition to two teaching hospitals and a research institutions,are institutions of higher learning,the highest volume of research institutions is the Beijing University of Traditional Chinese Medicine.Finally,the main content of the acupuncture experiment is to observe the mechanism of acupuncture intervention on the specific disease model.The acupuncture method is mainly electro-acupuncture,and the depth of the study has deepened to the level of gene expression.2)Simple acupuncture experimental study of high frequency selection points are mainly ST36(ZUSANLI),GV20(BAIHUI),GV14(DAZHUI),SP6(SANYINJIAO),GV26(SHUIGOU),PC6(NEIGUAN).BL23(SHENSHU),LI11(QUCHI).The high frequency effect indexes of the three levels of body function,micro function,tissue cell include learning and memory impairment,pain,neurological signs,neuronal discharges,blood pressure,heart rate,electrogastrogram,Cell apoptosis,cell count,infarct size,white blood cell count,body weight,etc.Analysis of acupuncture points and these indicators demonstrated that a point can be adjusted multiple effects indicators and an effect indicators can have multiple points to adjust the network relationship,the Analysis of this network relationship found a wide range of points to adjust their indicators are more,and the specific indicators have focused,reflecting the acupuncture experimental study of the hole when considering the specificity of the acupuncture points Its extensiveness,but also confirmed the feasibility of the calculation method in this study.3)Construction of acupuncture site-molecular relationship network.By calculating the complex network,the closed-type dense and complex network with complex network characteristics is obtained.Obtain the core acupuncture group that regulates the molecular index by calculating degree,closeness and betweenness of the network.Cluster analysis of this network to find a molecular interaction group associated with metabolic pathways containing five acupoints:ST36(ZUSANLI),GV26(SHUIGOU),PC6(NEIGUAN),ST34(LIANGQIU),ST2(SIBAI);and twelve genes:ATP5H,AK1,PAFAH1B2,PGAM1,UQCRFS1,PLCB1,COX5B,CKB,PRDX6,NME1,GAPDH,and FUT4.These five acupuncture points can be used to treat spleen and stomach diseases and related symptoms,suggesting that acupuncture these acupuncture treatment of spleen and stomach disease,the biological mechanism may achieve treatment Purpose by stimulating the 12 genes to regulate metabolic pathways,also speculated that foot Yangming stomach and metabolic pathways are closely related,which remains to be further studied to confirm.4)The main diseases of the acupuncture experimental literature include cerebral ischemia,Alzheimer's disease,spinal cord injury,vascular dementia,depression and other neurological diseases,and rheumatoid arthritis,obesity,gastric mucosal injury and other metabolic diseases,as well as myocardial ischemia as the representative of cardiovascular disease;Selection of high frequency of the acupoints are ST36(ZUSANLI),GV20(BAIHUI),PC6(NEIGUAN),GV14(DAZHUI),GV26(SHUIGOU),SP6(SANYINJIAO),BL23(SHENSHU),CV4(GUANYUAN),LI11(QUCHI),LI4(HEGU),etc.The relationship between the disease model and the acupuncture site was found to be complex.The results showed that GV20(BAIHUI),GV26(SHUIGOU)and GV14(DAZHUI)were focused on the treatment of brain diseases such as cerebral ischemia,cerebral ischemia-reperfusion injury and Alzheimer Disease;PC6(NEIGUAN)points are more focused on the treatment of heart disease.Combined with clinical applications,visible modern acupuncture animal experimental study is based on the basic theory of Chinese medicine to select the Acupuncture points.5)In the course of acupuncture experiments,the selection of acupoints in different stages of cerebral ischemia has the same focus on each.In the acute phase of cerebral ischemia,reperfusion injury,relative recovery period,sequelae of these four stages will be used to acupuncture points GV20(BAIHUI),ST36(ZUSANLI),GV14(DAZHUI),GV26(SHUIGOU),SP6(SANYINJIAO),PC6(NEIGUAN),LI4(HEGU).The effect of different stages in functional score,micro function and tissue cell level was basically the same,and the specificity was manifested in the microscopic function level.The relative recovery was related to the change of cerebral hypoxia.The spasticity was more concerned with the change of muscle tension,These two indicators were not detected in other periods;at the same time,most of the molecules of cerebral ischemic spasticity are neurotransmitters and their receptors associated with central spasm.It is reported that acupuncture can effectively regulate the expression of these molecules,and speculate that there is a close relationship between the mechanism of acupuncture in treating central spasm and the expression of these molecules,but the research on this aspect is relatively small.Therefore,acupuncture treatment of cerebral ischemia recovery and spasticity of the biological molecular mechanism there is still much room for exploration.
