The Study On Extraction,Purification And Pharmacodynamics Of Flavonoids In Dracocephalum Tanguticum Maxim

Dracocephalum tanguticum Maxim is one of the commonly used medicines in Tibetan areas.There have been reports about the constituents and pharmacologic action of Dracocephalum tanguticum Maxim since the 1990s.While no quality standards come into being so far.To expand the application scope of this premium crude medicine,here,we explore the quality standards of Dracocephalum tanguticum Maxim according to the Chinese Pharmacopoeia edition(2010)for better control of the medicine quality.Studies have shown the flavonoids serve as the main antiviral and antibacterial materials in terms of Dracocephalum tanguticum Maxim,while the liquid concoctions,a traditional treatment method,discount the medicinal effect in some degree.So it seems necessary for us to establish the extraction and purification system for further pharmacodynamic and pharmaceutical research in flavonoids,which provides a theoretical basis for further research and application.1,Actualized the determination of moisture,ash,extract of Dracocephalum tanguticum Maxim according to related stipulation of Chinese Pharmacopoeia.2,Established the HPLC,a stable and reliable methods of detecting luteolin-7-o-glucoside in Dracocephalum tanguticum Maxim.3,Summarized the factors affecting the extraction rate of Flavonoids according to the relevant literature and the actual production.Optimized the best extacting procedure of total flavonoids using orthogonal experiment method.4,Selected the appropriate D101 macroporous resin for purifying flavonoids through the static adsorption and desorption rate experiments;then examined the kinetics curves,the effect of different eluent concentrations,and the adsorption curves of different sample concentrations;finally concluded the optimum purification procedure.5,Antibacterial assays show that the flavonoids purification of Dracocephalum tanguticum Maxim is better than its water extract and alcohol extract,as well as honeysuckle particles on the bacteriostatic effect on Escherichia coli,Staphylococcus aureus and Pseudomonas aeruginosa.6,The in vitro antiviral experiments indicate that the extracts have no obvious effect on the cytopathy induced by virus,direct killing and prevention of virus infection.7,Carried out the pharmaceutical research in flavonoids purification of Dracocephalum tanguticum Maxim;determined the proportion of main medicine to other supplementary material in capsules;concluded the HCl-Mg reaction and TLC identification methods;established a method of determinating the content of main medicine;checked the capsule appearance,solubility,moisture and infilling differences,and the results show these aspacts can reach the standards of Pharmacopoeia.
