Licorice flavonoids have widely pharmacology action.The whitening effect of Licorice flavonoids is widely application in modern medicine and cosmetics industry.The reason of whitening is inhibit tyrosinase activity to inhibiting the synthesis of melanin,thus reduce the pigmentation production.The first part of this paper mainly use the licorice flavonoids content and the tyrosinase action as indexes.use polyamide column as separation conditions,examines the optimal extraction and purification process of total flavones.Make sure the specifications of polyamide?the pH?the best loading amount(the original solution of the extract from Glycyrrhiza uralensis Fisch.)?the concent ration of the sample solution?the desorption solvent?desorption velocity?elution volume.Through the experiment get the optimal purification process for separating the total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis Fisch was the best adsorption and desorption abilities of polyamide is 30?60 mech,the best pH of sample solution is at pH =6,the best loading amount is 32.48 mg(the original solution of the extract from Glycyrrhiza uralensis Fisch.):1 g 30?60 mech polyamide,the concent ration of the sample solution is 3.215 mg·mL-1,the desorption solvent is 70%ethanol,desorption velocity is 3 mL·min-1,and elution volume is 11mL 70%ethanol ·100 g-1 total flavonoids.trough verification tests to get use the select the optimal parameters that the yield is high,cost low,security and simple,avoid using organic solvents,suit for the big production.Also examines the macroporous resin separation licorice flavonoids process.Compared with polyamide when use macroporous resin to purification process for separating the total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis Fisch,the optimal purification process for separating the total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis Fisch was the best adsorption and desorption abilities of macroporous resin is sp825,the best pH of sample solution is at pH =3,the concent ration of the sample solution is 4.22 mg ·mL-1,the desorption solvent is 80%ethanol,desorption velocity is 2 mL·min-1,and elution volume is 15mL 80%ethanol ·100 g-1 total flavonoids.The results show that macroporous resin and polyamide resin could be used to separating the total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis Fisch.trough verification tests to get use both of select the optimal parameters that the yield is high,cost low,security and simple,avoid using organic solvents,suit for the big production.The second part of this paper discusses the effective parts of total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis Fisch.We according to different polarity of licorice flavones contains to separate.Extracted the licorice flavones in turn by petroleum ether,ethyl acetate,n-buoh.Determination the Tyrosinase inhibited action of part of petroleum ether,ethyl acetate,1-butanol,water soluble Were 35.3%,67.23%,40.36%,15.12%.Comparaed with the total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis,the Ethyl acetate part of the activity is the strongest.Through the active guidance and segmentation separation,we obtain the part of petroleum ether:Ethyl acetate(40:1),petroleum ether:Ethyl acetate(20:1),petroleum ether:Ethyl acetate(10:1),petroleum ether:Ethyl acetate(5:1),petroleum ether:Ethyl acetate(2:1),Ethyl acetate and methanol.Determine the part of tyrosinase inhibited activity.The results of tyrosinase inhibited activity were 2.14%,6.33%,30.38%,88.32%,81.61%,60.98%,48.89%,34.11%,23.79%..According to the experimental result of the above,we determined the total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis Fisch were exists in the part of petroleum ether:ethyl acetate(5:1),and petroleum ether:ethyl acetate(2:1).This research work was optimized purification process for separating the total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis Fisch,provides a research basis for product the total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis Fisch in industry..Also discusses the part of the total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis Fisch and basis for study of the part of total flavones that have Tyrosinase inhibitory action from Glycyrrhiza uralensis Fisch in the future.
