Swertia punicea is annual herb of Gentianaceae.It is distributed in Yunnan,Sichuan,Guizhou,western of Hubei and Hunan.It generally grows in the hillside meadow,flood,forest,thickets with altitude of 400-3800 meters.Its effects are liver gallbladder and dehumidifying heat.It is mainly used for jaundice and cholecystitis.Column chromatography and recrystallization were used for separation,NMR,UV and MS spectroscopic methods were used for structure identification,14 compounds were isolated from Swertia with 8 xanthone components,and respectively identified as:8-0-?-D-glucopyranosyl-1,5-dihydroxy-3-methoxyxanthone,Tetrahydrosertianolin,1,3,7,8-tetrahydroxyxanthone,1,3,8-three hydroxy-5-methoxyxanthone,1,5,8-trihydroxy-3-methoxyxanthone,1,7-two hydroxy-4,8-trimethoxyxanthone,2-D-glucopyranosy 1-1,3,6,7-tetrahydroxyxanthone,3-0-demethyl swertipunicoside.The above compounds are already isolated.Using Vc and BHT as positive controls,free radical scavenging ability of xanthones components from Swertia Puniceu was determined by DPPH method and IC50 of each compound was calculated.Scavenging ability of seven compounds had dose-effect relationship with concentration in a certain concentration range;scavenging ability orn DPPH radical of 1,3,7,8-tetrahydroxy xanthones,1,5,8-trihydroxy-3-methoxy xanthones,2-D-glucopyranosyl-1,3,6,7-tetrahydroxy xanthones were stron-ger than Vc;others are lower than Vc and BHT deriving from the IC50.HPLC was used for determination of five xanthones components from Swertia Punicea Hemsl.Method:Chromatographic column:AlltimalCl8(250mm×4.6mm,5(?m).Mobile phase meth-anol-water(including 0.06%H3PO4),and gradient elution.Flow rate:1 mL·min-1.Wavelength:254nm.Column temperature:30? The calibration curves of six xanthones components were in good linearity over the range of 14-140?g·mL-1,6-60?g·mL-1,4-40?g·mL-1,5-5?g·mL-1,9-90?g·mL-1,8-80?g·mL-1,(r=0.9996,0.9995,0.9997,0.9999,0.9996,0.9994).The average recoveries were99.72%,99.21%,98.92%,99.32%,98.80%,99.31%,RSD were less than 2%(n = 5).8-O-?-D-glucopyranosyl-1,5-dihydroxy-3-methoxyxathone was the highest concentrations in the whole plant Swertia Punicea.
