Systematic Separation Of Chemical Compositions Form Uncaria Rhynchophylla

Objective:The present study is to research the chemical constituents of Uncaria rhynchophylla,and identification the structure of the chemical components,also determination the content of triterpenoid,in order to provide experimental basis for future research.Method:Crushing herbs,extracted with 80% ethanol,ethanol recovery,then obtain extracting solution and slag after filter.Extracting solution extract successively with petroleum ether(60~90 ?),dichloromethane,ethyl acetate and n-butanol.Slag extract successively with petroleum ether(60~90 ?),dichloromethane,ethyl acetate and n-butanol.Every part were isolated and purified by the chromatography on repeated silica gel,macroporous resin,sephadex LH-20,TLC and HPLC,etc.The structure of the compounds was identified by 1HNMR and 13 CNMR.Use HPLC for determination of oleanolic acid and ursolic acid in Uncaria rhynchophylla content.Result:Twenty-four conpounds were isolated and purification from the Uncaria rhynchophylla in the end.They are two anthraquinones included: erythroglaucin(compound I)and physcion(compound II),two steroids included: sitosterol(compound III)and daucosterol(compound VIII),twoaliphatics included : palmitic acid(compound IV),tetracosane(compound XXIII),three triterpenes included: oleanic acid(compound V),ursolic acid(compound VI),?-amyrenol(compound VII),one steroids included: scopoletin(compound X),three phenolic acids include:ethyl 3,4-dihydroxy benzoate(compound XI),methyl rosmarinate(composund XII),ethyl caffeate(compound XIII),four flavonoids included: quercetin(compound IX),L-epicatechin(compound XIV),hyperoside(compound XVI),rutinum(compound XVII),seven alkaloids included:cadambine(compound XV),isorhynchophyllic acid(compound XVIII),rhynchophylline(compound XIV),geissoschizine methylether(compound XX),hirsutine(compound XXI),corynoxeine(compound XXII),harmane(compound XXIV).Oleanolic acid content in three samples respectively is 0.058 %,0.041 % and 0.049 %,ursolic acid content in three samples respectively is 0.64 %,0.42 % and 0.25 %.Conclusion:Through the separation of Uncaria chemical composition,twenty-four known compounds were isolated from the Uncaria,compounds I?IV?VII?XI?XII?XIII and XXIII were isolated from the Uncaria for the first time.To established the HPLC for the first time at the same time measure two triterpene compositions in Uncaria,in order to future study the chemical composition of Uncaria and provide experimental basis for quality standard.
