To morphologically and anatomically describe the gall of Quercus infectoria Oliv.and to describe the micromorphological characteristics of the insect galls and to provide scientific basis for identification the quality of the galls of Quercus infectoria,which are a kind of traditional Uygur medicine.To seperate and identify the chemical composition and structure of the gall of Q.infectoria.To establish a multi-components LC-MS fingerprint of the water extraction of the gall of Q.infectoria from different regions,and to determine the contents of Gallic acid and main components.Macroscopic identification,microscopic identification,physical and chemical identification,and LC-MS analysis were employed for the pharmacognostic study of the gall of Q.infectoria.GC-MS and FT-IR analysis methods were used to screen the chemical composition of the gall of Q.infectoria.LC-MS analysis technology was used to analyze and identify the components of the gall of Q.infectoria.A quantitative analysis of multi-components by single marker method was used to determine the content of main components,gallic acid was used as a reference.Similarity evaluation and hierarchical cluster analysis were employed to evaluate the gall of Q.infectoria..The galls of Q.infectoria were globular and had a short shank,the diameter was 1-2.5 cm and the thickness of wall was about 3-4 mm.The appearance was pale yellow and had verruca on the surface.The larvae had flown out,there was a hole connected to the surface,and the internal had larvae shell.The gall of Q.infectoria of imperforate had a larvae body,which was odorless and bitter.Transverse sections of the gall of Q.infectoria showed that the epidermal cells were square,the cortexes were wide with nonglandular hairs,and the cytoderms were thick and slightly lignified.Vascular bundles were collateral and numerous and scattered in parenchymal cells.Microscopic observation of the powder of the gall of Q.infectoria revealed sclereids that were either globular or rectangular and had a large cell cavity(5-10 ?m).Porous cells were densely distributed among the parenchyma cell walls and the vessels were mostly spiral.The gall of Q.infectoria mainly contained polyphenols,tannins,flavones.The maximum UV absorption wavelength of the gall of Q.infectoria were 212.5 nm and 270 nm.The results of LC-MS and MS analysis indicated that the gall of Q.infectoria contain gallotannins.GC–MS chromatogram of different extraction of the gall of Q.infectoria was established.8 representative peaks were identified as 3-(4-Isopropylphenyl)-1,1-dimethylurea,Ethyl iso-allocholate,Dibutyl Phthalate,(Z)-9-Octadecenoic acid 3-(octadecyloxy)propyl ester,6-Methyloctadecane,17-Pentatriacontene,8-(1,2-Dibromo-2-phenylethyl)-3,7-dihydro-1,3,7-trimethyl-1H-purine-2,6-dione,Methyl-11-((2R,3R)-3-pentyloxiran-2yl)undecanoate,respectively.The LC-MS fingerprint of the water extraction of the gall of Q.infectoria from different regions was obtained and got fourteen peaks,8 main peaks were identified as gallic acid,digallic acid,Monogalloyl-glucoside,Digalloylglucoside,Trigalloyl-glucoside,Tetragalloyl-glucoside,Pentagalloyl-glucoside,Hexagalloylglucoside,respectively.The similarity values of the eight batches of the gall of Q.infectoria from different regions were between 0.883 and 0.944.The characteristics of pharmacognosy could provide reliable basis for the identification of the gall of Q.infectoria medicinal materials.GC-MS and FT-IR analysis could quickly and accurately identify the main components of the gall of Q.infectoria.Furthermore,the establishment of LCMS fingerprint can determine the chemical compositions of the gall of Q.infectoria.This method is simple,rapid and has a high sensitivity and good repeatability.This work provide a reference method for quality standard of tannins-rich medicinal materials.
