Objectives: This study was designed to observe the changes of heat shock protein 60(HSP60)expression in the development of acute hepatic injury and analyze the relationship between HSP60 and acute hepatic injury of rats.Methods : 126 SD rats were randomly allocated into three groups: blank control group,non-liver injury group and liver injury group.The rats in control group without any treatment.The rats received treatment of left leg injury to fracture in non-liver injury group.Acute hepatic injury was induced by modified biological impact machine-IV(BIM-?)in liver injury group.Blood was respectively collected at 6hours,l2 hours,1thday,2nd day,3rd day,5thday,7th day to detect the alanine aminotransferase(ALT)and aspartate transaminase(AST)levels.At the same time,the samples were cut and the livers of rats were separated,and pathomorphological change of liver tissue samples were examined under optical microscope by hematoxylin-eosin(HE)stainning.The immunohistochemical staining was used to detect the expression changes of HSP60 during acute liver injury in rats.Results:The levels of ALT and AST in non-hepatic injury group did not significantly change compared with control group at each time point as mentioned above(P>0.05).The ALT and AST levels of hepatic injury groupincreased significantly at the 6 hours(104.17±4.07U/L,271.00±15.84U/L)and peaked at 12 hours(424.50±10.84U/L,787.70±32.68U/L),then went down and returned normal levels on the 3rd day.HE staining showed inflammatory cell infiltration and cell degeneration in hepatic injury group,the damage was most marked at the12 hour.The immunohistochemical staining showed that the levels of HSP60 in hepatic injury group at 6 hours was significantly increased and reached the highest at 12 hours compared with theblank control groups.The expression of HSP60 went down on the 1th,2th,3th day,but was still higher than the control group until returned basal level on5 th day.There was significant correlation between the level of HSP60 and ALT(r=0.9064,P<0.001)and AST(r=0.9294,P<0.001).Conclusions : The expression of HSP60 significantly changes during acute liver injury,indicating that the expression of HSP60 may play a protective role during alcoholic liver injury in rats.
