Objectives:To observe the effect of Chinese herbal compound formula Yizhi Congming Decoction(YCD)with the therapeutic principles of eliminating phlegm for resuscitation and improving intelligence on learning and memory capabilities of scopolamine model rats,and to explore its related mechanism.Methods:80 SD rats,half male and half female with the weight of 180-220 g,The behavioral tests(shuttle box test and Morris water maze)were applied to screen the rats with average intelligence and weed out the rats with relatively slow reaction,the last remaining 70 rats.70 rats were randomly divided into seven groups,namely,blank control group,model control group,positive-drug control group,Chinese herb control group,YCD group with low-,middle-and high-dose group.the rats in the blank control group and the model control group were given the same dose of normal saline.The positive-drug group was administrated with Piracetam Tablets(0.4g/kg).The Chinese herb control group was administrated with Guipi Pills(0.25g/kg).The gavage doses of the YCD low-dose group(10g/kg),the YCD middle-dose group(10g/kg),the YCD high-dose group(40g/kg)and the Chinese herb control group(20g/kg)were calculated according to weight.The administration lasted for 28 days.Behavioral tests were performed on the 23 nd day.Scopolamine injection was used to induce dysmnesia model.2.0 mg/kg of scopolamine injection was administrated to model control group,positive-drug control group,Chinese herb control group,YCD group with low-,middle-and high-dose group while normal saline was given to blank control group half an hour before the experiment.After administration,the behavioral tests(shuttle box test and Morris water maze)were performed.Then,SOD,GSH-Px and MDA were detected by biochemistry Elisa method.bax gene and bcl-2 gene were detected by q RT-PCR method.GFAP and BDNF were detected by immunohistochemical method(IHC).Results:1.According to the Morris water maze test and shuttle box test results showed that compared with the control group,the model group the average incubation period was longer than normal group(P < 0.05);rats unresponsive,shock time was significantly prolonged(P < 0.05),suggesting that memory impairment model successfully.2.After treatment,the average incubation period of positive-drug group and YCD middle-dose group were significantly shorter,which showed statistical difference(P <0.05);the Chinese herb group compared with the model group,the average incubation period was significantly reduced,there were significant differences(P < 0.05).The shuttle box test showed that the reaction of rats in YCD middle-dose group active,compared with the model group,shock time passive avoidance time is significantly reduced,there were significant differences(P<0.05).3.Compared to blank control group,the learning and memory abilities of model group were lower,SOD content was lower(P<0.05);MDA content was higher(P<0.05);GSH-Px activity was lower(P<0.05);Compared to model control group,the learning and memory abilities of YCD middle-and high-dose group were higher,SOD content was higher(P<0.05);MDA content was lower(P<0.05);GSH-Px activity was higher(P<0.05).4.Compared to blank control group,the learning and memory abilities of model group were lower,m RNA expression of Bax was higher(P<0.05);m RNA expression of Bcl-2 was lower(P<0.05);Compared to model control group,the learning and memory abilities of YCD middle-and high-dose group were higher,m RNA expression of Bax was lower(P<0.05);m RNA expression of Bcl-2 was higher(P<0.05).5.Compared to blank control group,the learning and memory abilities of model group were lower,the positive expression of GFAP was higher(P<0.05);the positive expression of BDNF was lower(P<0.05);Compared to model control group,the learning and memory abilities of YCD middle-and high-dose group were higher,the positive expression of GFAP was lower(P<0.05);the positive expression of BDNF was higher(P<0.05).Conclusion:1.The dysmnesia rat model induced by scopolamine injection is a reliable animal model,which has a more similar status of dysmnesia rats after medication.2.YCD could improve the learning and memory abilities of dysmnesia rats,its mechanism may be related to the increase of SOD content,decrease of MDA content,improvement of GSH-Px activity,relief of brain injury,decrease of free radical,less brain toxin and enhancement of antioxidant ability,etc.3.YCD could improve the learning and memory abilities of dysmnesia rats,its mechanism may also be related to the increased positive expression of BDNF and Bcl-2,decreased positive expression of GFAP and Bax,and reduced hippocampus apoptosis.
