Quantitative Determination Of Myocardial Marke RS With Fluorescence Immune Analysis

In recent years,acute myocardial infarction with high mortality and acute onset and other characteristics,has a significant threat to human health.Creatine kinase isoenzyme(CK-MB)and myoglobin(Myo)are important markers of myocardial injury in myocardial cells,and have important clinical significance in the diagnosis of acute myocardial infarction.In this study,the quantitative method of Myo and CK-MB in blood was established by using fluorescent microsphere with immunoassay analysis,and a quantitative test strip was also prepared.Principle is the combination of immunochromatography and photoluminescence.The monoclonal antibody and fluorescent microspheres were conjugated to a fluorescent label by EDC and NHS,and another antibody against the different target of antigen was coated onto NC membrane.Finally,sandwich structure consists of two kinds of monoclonal antibodies and the antigen in the sample.The excitation wavelength of the fluorescent microspheres was 365 nm,and red light was emitted under ultraviolet light.The relative fluorescence intensity was measured by a fluorescence quantitative detector to quantitatively detect the content of Myo and CK-MB.In this paper,I have done a series of experiments,such as fluorescent microsphere labeling and characterization,screening of raw materials,immunochromatographic test strips to build and optimize methodological assessment.Results: The linear range of Myo test strip was 25 ng / ml ~ 1500 ng / ml,the level of sensitivity was 12.5 ng / ml,CV <7%.The linear range of CK-MB test strip was 1 ng / ml ~ 1000 ng / ml,the sensitivity was 1ng / ml,CV <8%.Sample test results showed thatfluorescent immunochromatographic test strips and the purchase of colloidal gold test strip has a good correlation.But the fluorescent test strips can be quantitative detection of samples,colloidal gold can only qualitative detection,and fluorescent test strips more sensitive.The experimental results provide experimental basis for the development of Myo and CK-MB fluorescence detection method and detection card.
