Object:To investigate the expression of CD133,Nestin and Sox2 in meningiomas and explore whether braint tumor stem cells exist in meningiomas.in order to make clear whether exist in different level meningioma tissue brain tumor stem cells,and carries on the comparison to these markers positive rate,offers a new target for the diagnosis and treatment of meningioma.Methods:1.Gather specimens after surgical resection in 35 cases of meningioma,And grouping: There are 6 males,and 29 females;The average age was 52.49±11.55 and grouping: pathological results WHO grade I specimens for benign group 28 cases;WHO II level III specimens for 7 cases of malignant group(5 cases to WHO ?,2cases to WHO ? level).Collected samples were obtained with 10% formalin and liquid wax packages,for cutting.2.Using immunohistochemical techniques to detect CD133,Nestin,SOX2 expressed in 35 cases of meningioma in the organization,after staining,microscope observation and collect pictures and images to make use of IPP software analysis,measurement of positive cells of total Optical density(Integrated Optical Desity,IOD)values.And the expression of each marker in statistical analysis.Results:1.CD133 are mainly distributed in meningioma tumor cells and vascular endothelial cell membrane and cytoplasm,positive cells were brown;Nestin are mainly distributed in meningioma of tumor cells and vascular endothelial cell cytoplasm,the positive cells were brown;The SOX2 are mainly distributed in meningioma tumor cells and vascular endothelial cell membrane and cytoplasm,positive cells were brown.2.CD133,Nestin in different levels meningioma tissues are expressed,and the expression level increased as the pathological level increases;The SOX2 expressed in malignant meningioma group,and no expression in benign group.3.At different levels in meningioma CD133,Nestin,SOX2 is respectively:CD133benign group IOD = 33251 ± 9562,CD133 positive cells and malignant groups IOD= 139745 ± 26775;Benign group of Nestin positive cells IOD= 22389±7031,Nestin positive cells and malignant group IOD= 255707±39204;Benign group SOX2 IOD = 0,SOX2 positive cells and malignant groups IOD = 681284±46372.At different levels in meningioma CD133,Nestin,SOX2 positive cells of IOD values have significant difference(P < 0.05).Conclusion:The results showed that the tumor stem cell marker CD133,Nestin,SOX2 expression in human meningioma tissue showed that existing in meningioma of cancer stem cells,and the expression level of positive cells and tumor is closely relative to the malignant degree,can detect the index judging malignant degree and prognosis of tumor,and with the occurrence of meningioma,development,closely related to the outcome.In the diagnosis of meningioma,prognostic and search for new therapeutic targets is of great significance.
