Objective:To study the characteristics of CD133-positive and Nestin-positive human glioma stem cells proliferation index(PI) and investigate the colocalization and relationship of CD133-positive and Nestin-positive human brain glioma cells.Methods:①Immunohistochemistry(IHC) was used to detect the expression of CD133 and Nestin in human brain glioma tissue.②Double staining immunohistochemistry was used to detect co-expression of CD133 and Ki-67 as well as co-expression of Nestin and Ki-67 in human brain gliomas.Results:①We found that the PI of CD133-positive cells in low-grade gliomas was lower than that in high-grade[(14%,8%~66%) vs(93%,87%~95%),P=0.001];the PI of Nestin-positive cells in low-grade gliomas was higher than that in high-grade[(28.5%,21%~46%)vs(15%,8%~22.5%), P=0.019];the PI between CD133-positive and Nestin-positive cells in low-grade gliomas had no statistically significant difference[(14%, 8%~66%) vs(28.5%,21%~46%),P=0.395),but the PI of CD133-positive cells was higher than that of Nestin-positive cells in high-grade gliomas(93%,87%~95%) vs(15%,8%~22.5%),P=0.000];the percentage of proliferating CD133-positive cells in proliferating glioma cells was higher than that of proliferating Nestin-positive cells in proliferating glioma cells in low- and high-grade gliomas,respectively[(83.5%, 61.5%~91%) vs(22.5%,10%~48.5%),P=0.004;(53%,37%~65%)vs(24%, 10.5%~42.5%),P=0.003].②CD133 and Nestin were expressed in different cell types in the same glioma tissue.Conclusions:①This study suggests that PI of CD133-positive glioma cells increases as the glioma malignancy increases.The PI of Nestin-positive glioma cells decreases as the glioma malignancy increases. CD133-positive cells are the main proliferating cells in tumor tissues. ②CD133-positive cells and Nestin-positive cells may be the different cell subtypes.
