Experimental Study On The Effects Of P53?PUMA Protein With The Lizard Weikang Basic Prescription And The Different Combination On Precancerous Lesion Of Gastric Cancer In Model SD Rats

Objective The objective of this study was to explore the Lizards weikang basic prescription,tonic prescriptions of Qi group,nourishing the Yin group,relieving toxicity and dredging meridians group on the gastric mucosa cell proliferation,apoptosis and P53 level of PLGC model rats.Furthermore,its mechanism of action on PLGC was expounded,the clinical application of the prescription to provide the basis for treatment of PLGC.Methods 70 SPF,SD male rats,4 to 6 weeks,were randomly divided into blank control group(A),PLGC in model control group(B),tonic prescriptions of Qi group(C),nourishing the Yin group(D),relieving toxicity and dredging meridians group(E),the Lizards weikang basic prescription group(F),Vitacoenzyme treatment group(G).There are altogether seven groups in this experimental,and 10 rats in each group.In addition to the blank control group feeding conventionally.Other groups use 0.02mol/LN-methyl-N'-nitro-N-nitrosoguanidine(MNNG)and 40% ethanol to lavage,and also can drink MNNG freely.Furthermore,it include hunger disturbance(2 d satiety,ld hunger)comprehensive factors.In the aggregate of 24 weeks to building the model for the PLGC of rat.After 24 weeks,1 rat randomly chosen from each group,and kill them to confirm the pathological diagnosis criteria of PLGC.After make model building successfully,through the pathological detection,the blank control group and PLGC in model control group were given saline lavage,while other groups were given corresponding lavage treatment drugs.All of them were given drugs 2 times/day and for 12 continuous weeks.Observe the general condition of each group,and at the end of 12 th week,snapping the rats fasting the water for 24 h,then narcotize and put to death the rates.After this process,observed the histopathological changes of gastric mucosa,and the routine HE staining was used to observe the histopathology.Detect the factor P53 and PUMA expression by immunohistochemicl methd.At last,statistical data were used to analyze the experimental data.Results1.General situation: blank control group of rats is obesity,very sensitive,and in good spirits.Their fur are bright,faeces are normal,and their diet are as usual.PLGC in model control group of rats is emaciation.These rats activities less,listlessness,fur bleak,eating water reduced,soft stools.Each treatment groups are generally in good condition,and the above performance is better than that of PLGC in model control group.2.Histopathological observation: Found by naked eye,blank control group of gastric folds shape normal,the shape and color are normal,the pink.Mucous membrane of gastric mucosa is smooth,and mucus is more.The PLGC in model control group gastric folds disorder,the bleeding points and erosion in scattered,the dark white,and gastric mucosal surface is not smooth.Each treatment groups are generally in good condition.The recovery of gastric mucosa of the Lizards weikang basic prescription group was the most obvious.Found by the microscopic,blank control group of gastric folds' structure is normal.The PLGC in model control group gastric folds are different degrees of atrophy and intestinal metaplasia and dysplasia.Each treatment groups on the gastric mucosa's atrophy and intestinal metaplasia and dysplasia was improved in different degrees,most of all,the Lizards weikang basic prescription group improved significantly.3.Immunohistochemical results: Comparison between PLGC in model control group,Each treatment groups and blank control group,(P(27)0.05);Comparison between each treatment groups and PLGC in model control group,(P(27)0.05);Comparison of tonicprescriptions of Qi group,nourishing the Yin group,relieving toxicity and dredging meridians group and he Lizards weikang basic prescription group,(P(27)0.05);Among them,the lowest AOD value in the Lizards weikang basic prescription group,the results showed that the treatment effect of this group was the most obvious;Comparison between tonic prescriptions of Qi group,nourishing the Yin group and relieving toxicity and dredging meridians group,(P(27)0.05);and the lowest AOD value in tonic prescriptions of Qi group,the results showed that the treatment effect of this group is better;The tonic prescriptions of Qi group,nourishing the Yin group and relieving toxicity and dredging meridians group compared with Vitacoenzyme treatment group,(P(27)0.05).Conclusion1.The Lizards weikang basic prescription and the different combination can improve the body weight change,activity and survival,mental state,hair color,intake of water,stool,and other general situation in rats with precancerous lesions of gastric cancer.2.The Lizards weikang basic prescription and the different combination can improve gastric precancerous lesion in a rat model of gastric mucosa gland atrophy,intestinal metaplasia and dysplasia.3.The Lizards weikang basic prescription and the different combination can influence the expression of P53 and PUMA protein in precancerous lesions of gastric cancer.4.Among the different combination of Lizards weikang basic prescription,the tonic prescriptions of Qi group is better than nourishing the Yin group and dredging meridians group in the repair of gastric mucosal injury.
