Platycodon grandiflorum A.DC is a species of pernnial flowering plant of the family Campanulaceae.Playcodi Radix?PR?,the dried root of Platycodon grandiflorum has been used as a troditional medicine in China.As a traditional medicine,PR can effectively treat coigh,abscess andexcessive phlegm with bitter,acrid and flat.Studies have shown that Platycodon grandiflorum contains triterpenoid saponins,polysaccharides,flavonoids,polyynes,steroids,phenolic acids,fatty acids and other types of compounds.The principal bioactive constiyuents of this herb are triterpenoid saponins,which includes Platycodin S,C,D,D2,D3,E.Among them,Platycodin D,is a triterpenoid saponin extracted from Platycodon grandiflorum,is the most widely used main active substance.In this paper,the extraction of saponins from the stem and leaf part and the root part of Platycodon grandiflorum was studied.The mechanism of pharmacological action was further studied,which was of great significance to the development of health care products and clinical therapeutic drugs of Platycodon grandiflorum.In this paper,the stem and leaves of Platycodon grandiflorum and the roots of Platycodon grandiflorum were extracted and separated.The stem and leaves of the Platycodon grandiflorum were crushed and the ethanol was added and extracted by ultrasonic extraction at 40 ° C for 30 min.Then extractive was collected extractive,with vacuum recovery of dry solvent to obtain a dry powder.Add distilled water to the maximum extent possible to dissolve it as a sample.After the D101 macroporous resin column was adsorbed for 12 h,the eluate was collected in a column volume of 10% with 70% ethanol column.The ethanol-obtained aqueous solution was freeze-dried.To obtain platycodin of stems and leaves.And used as samples and drugs for later experiments.The chemical constituents of saponins from Platycodon grandiflorum were analyzed by HPLC.Detection of its contains platycodin D,lobetyolin and other components;The Platycodon grandiflorum was crushed and extracted three times then ultrasonic extraction.The rotary evaporator was recovered under reduced pressure and evaporated to dryness,and the residue was washed twice with water,and the mixture was recovered under reduced pressure to obtain an alcohol.The powder was freeze-dried to obtain a powder as a late sample of the experiment and the use of drugs.The chemical constituents of Platycodon grandiflorum were analyzed by HPLC,and the contents of platycodin D,d-platycodin D and lobetyolin were detected,which laid the foundation for the future research and development of food and medicine.Previous studies have shown that stems and leaves of Platycodon grandiflorum are rich in saponins,flavonoids,polyphenols and so on.And has a strong antioxidant activity.In order to make full use of medicinal resources,the chemical composition and pharmacological activity of the stem and leaf of Platycodon grandiflorum should be studied in order to provide the necessary theoretical basis for future medicine.In this study,we used the conventional extraction and isolation technology to enrich the saponins from the stems and leaves of Platycodon grandiflorum?PSSL?,and establish the animal model of H22 transplanted tumor.The first observation of the anti-tumor effect of PSSL on mice.And its molecular mechanism to do a preliminary discussion.The results showed that compared with the model group,the quality of tumor H22 transplanted tumor in PSSL group?25 and 50 mg / kg?was decreased and the growth was inhibited.Liver,spleen and thymus index were close to the model group,showing that PSSL had protective effect on immune organs.The ALT and AST indices were decreased in each dose group of PSSL.Serum levels of TNF-? and IFN-? were significantly increased.In addition,H&E and Hoechst 33258 staining showed that PSSL could induce apoptosis of tumor cells.The above results indicate that PSSL is dose-dependent in H22 tumor-bearing mice in a dose-dependent manner,and the underlying mechanism is that PSSL improves immune function and induces apoptosis.Acetaminophen?APAP?is the most commonly used analgesics and antipyretic drugs around the world.Although it is safe at a given therapeutic dose,however,excess APAP often causes severe liver damage and liver failure.A previous study showed that Platycodin can significantly improve oxidative stress and inflammatory response to protect the liver.We established a model of acute liver injury induced by Acetaminophen?APAP?,to observe the protective effect of the root of Platycodon grandiflorum?PS?against on liver injury.The results showed that compared with APAP model group?250mg / Kg?,the high dose of PS?30mg / Kg?mice was significantly improved and the body weight was increased,which could significantly improve the liver index caused by APAP intoxication.The levels of serum ALT and AST,TNF-? and IL-1? were significantly decreased by administration of PS,and the content of MDA in liver tissue was decreased,whereas the content of GSH in liver tissue was increased in a dose-dependant manner.Histological analysis showed that PS could attenuate APAP-induced liver injury.The levels of TNF-? and IL-1? in the liver were measured by real-time fluorescence quantitative PCR.The expression of TNF-? and IL-1? was significantly increased after APAP administration,and PS treatment attenteand this elevation.In summary,PS has a protective effect on acute APAP-induced liver injury.In summary,this study shows that PSSL has anti-tumor effect on H22 tumor-bearing mice.Meanwhile,PS has beneficial action against APAP hepatic injury.Its possible mechanism is the improment of improving oxidative stress,regulation immune function,reduction the inflammatory response,inhibition lipid peroxidation and anti-apoptosis.
