Objective:Combing the records of Platycodon grandiflorum in ancient herbal books.Based on the records of"south Platycodon grandiflorum with good quality"and"bitter Platycodon grandiflorum with good quality",the samples of wild platycodon grandiflorum were collected widely,and the quality of Platycodon grandiflorum from different places were analyzed.the taste information of Platycodon grandiflorum from different places were studied to explore the scientific connotation and material basis of"south Platycodon grandiflorum with good quality"and"bitter Platycodon grandiflorum with good quality".Methods:1:Ancient herbal books such as"Shen Nong's herbal classic","Annotated Shen Nong's Herbal","classified Materia Medica",etc.were consulted to comb the germplasm,harvesting and processing,producing area evolution,quality evaluation and other related content of Platycodon grandiflorum.2:The total saponins contents of Platycodon grandiflorum from different places were studiedbyvanillin-glacialaceticacidandperchloricacidcolorimetric spectrophotometry;Establishing a HPLC-ELSD method for simultaneous determination ofDeapioplatycosideE,PlatycosideE,DeapioplatycodinD3,PlatycodinD3,Deapioplatycodin D,platycodinD2,PlatycodinD,PolygalacinD and combined with principal component analysis and cluster analysis to study the samples from different places.3:The characteristic fingerprints of Platycodon grandiflorum from different habitats was established and combined with principal component analysis and cluster analysis to study the samples from different places.4:The taste information of Platycodon grandiflorum from different points was detected by SA-402B taste analysis system,and the differences of taste information of Platycodon grandiflorum from north to south of China were investigated.Results:1:The records of Platycodon grandiflorum in ancient herbal books were mainly related to germplasm,harvesting and processing,producing area evolution,quality evaluation and so on.Among them,the medical workers in the past dynasties agreed the good quality of Platycodon grandiflorum with bitter taste and producing in southern regions.2:The study on the saponins from Platycodon grandiflorum showed that the total saponins contents of the wild Platycodon grandiflorum from 23 points was in the range of 9.584mg/g27.005mg/g,the average contents of samples from Lianchen town in Guangnan County,Yunnan,Tongcheng City,Anhui and Jinzhai County,were higher,the average contents of samples from Cangyanshan town in Jingxing County,Hebei,Qingta Village in She County,Hebei,Taohuatan town in Jing County,Anhui was lower.The contents of 8 kinds of saponins were higher in Jiangxi,Guizhou,Guangxi and Yunnan,were higher content of platycoinD.It was found that these Platycodon grandiflorum with higher saponins were located in south and southwestern regions of China.3:The fingerprints of Platycodon grandiflorum from 23 points were established by HPLC-UV method and 19 common peaks were identified.The similarity of Platycodon grandiflorum from different points was between 0.6070.921 and showed that the difference in composition between different points was large.4:The sweet,bitter and fresh taste information of Platycodon grandiflorum from different points were richer.The sweet taste of Platycodon grandiflorum in different provinces decreased from north to south,and the sweet taste of Platycodon grandiflorum in the north was higher than that of southern Platycodon grandiflorum.The bitterness and fresh taste of Platycodon grandiflorum in different provinces showed an increasing trend from the central regions to the south and the north.Conclusion:Through the herbal research of Platycodon grandiflorum,the records of the harvesting and processing,germplasm,producing area evolution,quality evaluation of Platycodon grandiflorum were clarified.The determination methods of the total saponins and each monomer of saponins of Platycodon grandiflorum were accurate and reliable,which can be used to evaluate the quality of Platycodon grandiflorum from different points.Researches showed that the south Platycodon grandiflorum with good quality mainly because of its higher saponins contents.The study of taste information of Platycodon grandiflorum from different points showed that the relationship between bitter taste and saponins contents was not obvious,but bitterness ratio was consistent with saponins.The literature recorded"northern Platycodon grandiflorum with sweet taste"was consistent with the experimental results,however,it's still unclear what kind of substance did the different taste of Platycodon grandiflorum produce from,so further study after subdividing and purifying the components of Platycodon grandiflorum should be conducted.
