Objective:Minichromosome maintenance protein 2(MCM2)and Survivin can promote the proliferation and inhibit the apoptosis of cervical cancer cells,so both play a role in the progression of cervical cancer.To investigate the effect of NACT on the expression of MCM2 and Survivin before and after neoadjuvant chemotherapy(NACT)in patients with locally advanced cervical cancer.To investigate the correlation between MCM2 and Survivin and the correlation with the efficacy of NACT in the treatment of locally advanced cervical cancer.The aim of this study was to predict the effect of NACT on the efficacy of NACT.Methods:Selected in March 2015-July 2016,Shanxi Medical University,Second Hospital gynecological outpatient colposcopy biopsy diagnosed as local advanced cervical cancer in 30 patients.Colposcopy with biopsy forceps clamp cervical cancer tissue,washed with0.9% Nacl tissue blood stains,aluminum foil wrapped,and stored in liquid nitrogen tank.All patients were treated with no contraindications,and had not received surgery and radiotherapy,in the case of patients signed informed consent to give two courses of TC program(paclitaxel + carboplatin)chemotherapy,cervical cancer after radical surgery Take the corresponding patients with fresh cervical cancer tissue,stored in liquid nitrogen cans.The total RNA and total protein of cervical cancer tissues were extracted and the expression of MCM2 and Survivin mRNA and protein were detected by Real-time qPCR and Western blot before and after NACT.Results:1.Local advanced cervical cancer patients after NACT tumor diameter was significantly reduced,by chemotherapy before(42.43 ±5.90)mm Shrink to(24.29 ±1.60)mm(P<0.05).The total effective rate was 83.3%.2.The expression of MCM2 and Survivin in patients with locally advanced cervical cancer after NACT was lower than that before chemotherapy.RT-qPCR detection the mRNA expression of MCM2 and Survivin after chemotherapy was 0.62±0.11 and 0.86± 0.13,respectively,which were significantly lower than that of pre-chemotherapy protein mRNA 1.31±0.23,1.28±0.19(P<0.05).Western blot analysis showed that the relative expression of MCM2 and Survivin after chemotherapy was 0.18±0.02,0.19±0.02,which was significantly lower than that of pre-chemotherapy protein 0.90±0.02,0.49±0.03(P<0.05).3.The expression of MCM2 and Survivin in the patients with locally advanced cervical cancer before NACT was higher than that in the ineffective group.RT-qPCR detection the relative expression of MCM2 and Survivin mRNA was 1.52±0.26,1.45±0.21,compared with the ineffective group,the expression of mRNA was 0.40±0.06 and0.40 ± 0.03(P<0.05).Western blot analysis the relative expression of MCM2 and Survivin in the effective group was 0.94 ± 0.01,0.52 ± 0.02,compared with the ineffective group,the expression of protein was 0.75±0.02,0.34±0.01(P<0.05).4.The expression of MCM2 and Survivin in locally advanced cervical cancer before NACT was found to be positively correlated by Pearson's product moment correlation,and the correlation coefficient r was 0.876.Conclusions:1.The expression level of MCM2 and Survivin in patients with locally advanced cervical cancer was significantly lower than that before chemotherapy,suggesting that it could be used as an effective screening index for assessing and predicting the effect of NACT on cervical cancer patients.2.MCM2 ? Survivin was highly expressed in the efficacy of locally advanced cervical cancer,suggesting that the degree of expression was helpful in assessing the patient's sensitivity to NACT.
