Expression And Clinical Significance Of Prion Protein In Laryngeal Squamous Cell Carcinoma

Objective:To observe the expression of prion protein(Pr Pc)in laryngeal squamous cell carcinomas(abbreviated as squamous cell carcinomas)and to explore the relationship between Pr Pc expression and clinical pathologic features of laryngeal squamous cell carcinoma.Methods:Application of Immunohistochemistry(Envision)assay to detect the expression situation of Pr Pc in 60 cases of laryngeal squamous cell carcinoma,in 20 cases of laryngeal squamous atypical hyperplasia tissue and in 20 cases normal laryngeal mucosa taken from patients without tumor.with each successive slice thickness 4?m.Making tissue microarray,Use provided by the Wuhan Institute of Virology,Chinese Academy of virology laboratory Chaoyang-Li 4h2 mouse anti-human prion protein monoclonal antibody as an anti-1.Related kit was purchased from Beijing Zhongshan Golden Bridge Biotechnology.in order to analyze the relationship between the expression level of Pr Pc and clinical pathological features of patients with laryngeal squamous cell carcinoma,such as,gender,age,clinical stages and pathological cell differentiation,and with or without lymph node metastasis.Results:The positive expression rate of Pr Pc in laryngeal squamous cell carcinomas organization,laryngeal squamous atypical hyperplasia tissue and normal laryngeal mucosa was 55%(33/60),25%(5/20)and 5%(1/20)respectively.There were statistically significant differences in expression of 3 kinds of tissues(P<0.05).With the decrease of pathological differentiation degree,the expression of Pr Pc in laryngeal squamous cell carcinomas organization rises(P<0.05).The positive expression rate of Pr Pc in laryngeal squamous cell carcinoma tissues of the advanced stage patients group(III stage and IV stage)was significantly higher than that of laryngeal squamous cell carcinoma tissues of I and II stage patients group(P<0.05).Conclusions: Pr Pc expression in a high state of squamous cell carcinomas is likely to participate in local invasion and metastasis of squamous cell carcinomas tissue and molecular pathways of distant metastasis,and bear important physiological functions in the process of tumor cells,and Pr Pc expression and TNM stage,predicting distant metastasis also has important clinical significance.It has been confirmed is closely related to the development of HPV virus infection and squamous cell carcinomas,the expression of Pr Pc in squamous carcinomas cells is affected by HPV virus infection needs further study,obviously Pr Pc expression in malignant tumors,which in tumor cells whether he will take the anti-apoptotic effect and is widely present in the cytoplasm of whether to undertake certain functions of tumor cell growth,pending further study.
