Objective: To explore the expression of microRNA-301 b and micro RNA-376 a in colorectal cancer and the adjacent tumor free normal mucosa tissues,and analyze the relationship between expression of microRNA-301 b and microRNA-376 a and clinic pathological features.Method:1 28 colorectal cancers and the adjacent tumor free normal mucosa tissues were obtained from Tangshan gong ren hospital;2 Total RNA from colorectal cancers and the adjacent tumor free normal mucosa tissues was extracted and reverse transcribed to cDNA.Real time PCR was used to detect the expression of miR-301b/ miR-376a;3 Relationship between relative expression of miR-301b/miR-376 a and clinic pathological features(gender,age,tumor location,invasive depth,differentiation,histological types,lymph node metastasis,distant metastasis,CEA,CA199,CA724)of patient was analyzed.4 GraphPad Prism 5 software was used for statistical analysis,all the data was represented as median(interquartile range),Wilcoxon test was used to analyze the data between two groups and P< 0.05 represents statistical significance.Result:1 Expression of miR-301 b in colorectal cancerThe relative expression of miR-301 b in colorectal cancer and the adjacent tumor free normal mucosa tissues was calculated by 2-Delta Ct method with the CT value of tumor free tissue as control.Medians of miR-301 b relative Expression in colorectal cancer and tumor free normal mucosa tissues were 6.564 and 0.9353(P=0.0061),with the interquartile ranges were(1.261,29.86)and(0.6053,3.180)respectively.There was a significant increase in miR-301 b expression in colorectal cancers.2 Analysis of relationship between miR-301 b and clinic pathological features.By using the method of 2-Delta Ct to calculate the miR-301 b relative expression,the relative expression of miR-301 b in each sample was evaluated by the ratio of Fc value in colorectal cancer divide to tumor free normal mucosa tissues.Expression of miR-301 b in well+medium differentiation and worse differentiation colorectal cancer were 1.383 and 57.31(P=0.0115)with the interquartile ranges were(43.44,126.9)and(1.022,11.45)respectively;Expression of miR-301 b in patients without distant metastasis and with distant metastasis were 1.248 and 12.43(P=0.0159)with the interquartile ranges were(0.6004,17.34)and(4.771,120.3)respectively;Expression of miR-301 b in patients with CA724 more than 5 and CA724 less than 5 were 1.392 and 126.9(P=0.0142)with the interquartile ranges were(1.056,14.89)and(42.46,146.3)respectively.3 Expression of miR-376 a in colorectal cancerThe relative expression of miR-376 a in colorectal cancer and the adjacent tumor free normal mucosa tissues was calculated by 2-Delta Ct method with the Ct value of tumor free tissue as control.Medians of miR-376 a expression in colorectal cancer and tumor free normal mucosa tissues were 1.685 and 1.164(P=0.0153)with the interquartile ranges were(1.079,2.332)and(0.326,1.966)respectively.There was a significant decrease in miR-376 a expression in colorectal cancers.4 Analysis of relationship between miR-376 a and clinic pathological featuresBy using the method of 2-Delta Ct to calculate the miR-376 a relative expression,the relative expression of miR-376 a in each sample was evaluated by the ratio of Fc value in colorectal cancer divide to tumor free normal mucosa tissues.Expression of miR-376 a in patients without lymph node metastasis and with lymph node metastasis were 1.248 and 0.586(P=0.0232)with the interquartile ranges were(0.717,2.025)(0.184,0.915)respectively;Expression of miR-376 a in patients without distant metastasis and with distant metastasis colorectal cancer were 1.070 and 0.522(P=0.0486)with the interquartile ranges were(0.527,1.727)(0.192,0.689)respectively;Expression of 376 a in patients with CEA more than 20 and CEA less than 20 were 1.032 and 0.464(P=0.0404)with the interquartile ranges were(0.773,1.715)and(0.188,0.571)respectively.Conclusion:1 As a cancer promoter,miR-301 b promotes formation of colorectal cancers and the invasion of colorectal cancer.2 As a cancer inhibitor,miR-376 a inhibits formation of colorectal cancers and metastasis of colorectal cancer.
