Objectives:The expression of MAGE-A3,MAGE-A9 protein and gene was detected by histopathological abnormalities in cervical lesions and compared with normal tissues.To study the relationship between MAGE-A3 and MAGE-A9 and the development and development of cervical intraepithelial neoplasia and cervical cancer,so as to provide a theoretical basis for the diagnosis and treatment of cervical lesions,and then to develop a practical treatment program.Methods:(CIN ?)40 cases,cervical cancer Ia ~ b period(FIGO staging)60 cases,cervical cancer IIa period(FIGO staging)24 cases,cervical intraepithelial neoplasia(CIN ?)Normal cervix in 40 cases.All patients underwent surgical treatment,access to patient data,combined with the patient's clinical and pathological data for analysis.The expression of MAGE-A3 and MAGE-A9 was observed under light microscope.The expression of MAGE-A3 and MAGE-A9 was observed by immunohistochemical staining.The expression of MAGE-A3 and MAGE-A9 gene in 15 cases of normal cervical,15 cases of CIN? and 20 cases of cervical cancer were detected by real-time fluorescent PCR.Happening.The expression of MAGE-A3 and MAGE-A9 protein in 15 cases of normal cervical,15 cases of CIN? and 20 cases of cervical cancer were detected by Western Blot.Result: 1.Immunohistochemistry showed that MAGE-A3 protein was positive in cervical cancer and cervical intraepithelial neoplasia,mainly located on the cytoplasm,and the expression level of MAGE-A3 protein increased gradually with the severity of cervical lesions.The positive expression rates of MAGE-A3 in normal cervix,CIN? and cervical cancer were 5.00%,15.00% and 48.81% respectively,and the positive expression of MAGE-A3 was gradually increased.The statistical analysis showed that the normal group and CIN? group(P <0.01).On the contrary,there was no significant difference between the normal group and the CIN? group(P> 0.05).The results showed that the expression of MAGE-A3 was significantly higher than that of the normal group(P <0.01)The expression level of MAGE-A3 was correlated with the clinical stage,pathological grade,lymphatic metastasis and HPV infection of cervical cancer(P <0.05).MAGE-A9 protein was positive in cervical cancer and CIN tissues.The positive expression rates of MAGE-A9 in normal cervical,CIN? and cervical cancer tissues were 7.50%,17.50% and 55.95% respectively,and the positive expression of MAGE-A9(P <0.01).Compared with CIN ? group,the expression of MAGE-A9 in normal group and CIN ? group was significantly higher than that in CIN ? group(P <0.01).(P <0.05).The results showed that the expression level of MAGE-A9 was correlated with the clinical stage,pathological grade and lymphatic metastasis of cervical cancer(p <0.05).2.Real-time quantitative PCR showed that:(1)The relative expression levels of MAGE-A3 mRNA in normal cervix,CIN? and cervical cancer tissues were 0.184 ± 0.069,0.221 ± 0.086 and 0.702 ± 0.145 respectively with GAPDH mRNA as reference standard.The relative expression of MAGE-A3 mRNA in cervical cancer was significantly higher than that in CIN and normal tissues.There was significant difference between cervical cancer group and normal group and CIN? group(P <0.05),but there was significant difference between normal group and CIN ? group No statistically significant(P> 0.05)(2)GAPDH mRNA as the internal reference standard,normal cervical,CIN? and cervical cancerThe expression level of MAGE-A9 mRNA in cervical cancer tissues was significantly higher than that in CIN and normal tissues,and the relative expression levels of MAGE-A9 mRNA were 0.182 ± 0.062,0.234 ± 0.087 and 0.642 ± 0.150 respectively.(P <0.05),but there was no significant difference between the normal group and CIN? group(P> 0.05),but there was no significant difference between the two groups(P> 0.05)3.Western bloting protein blotting shows:(1)The gray level of MAGE-A3 gradually increased with the severity of cervical tissue lesions.The relative expression levels of MAGE-A3 protein in normal cervix,CIN? and cervical cancer tissues were 0.943±0.178?1.035±0.139?1.977±0.105 respectively.The relative expression of MAGE-A9 protein in cervical cancer tissues was significantly higher than that in CIN? and normal cervical tissues(P <0.05),but there was no significant difference between normal group and CIN ? group(P> 0.05).(2)The gray level of MAGE-A9 gradually increased with the severity of cervical lesions,and the relative expression levels of MAGE-A9 protein in normal cervical,CIN and cervical cancer tissues were 0.857 ± 0.315,0.909 ± 0.526,1.882 ± 0.179,respectively.The relative expression of MAGE-A9 protein in cervical cancer tissues was significantly higher than that in CIN? and normal cervical tissues(P <0.05),but there was no significant difference between normal group and CIN ? group(P> 0.05).Conclusion:1.MAGE-A3 overexpression is closely related to the development of cervical cancer,and the overexpression of MAGE-A3 is closely related to the clinical stage,pathological grade and lymphatic metastasis and high-risk HPV infection of cervical cancer,but not with age and cancer type Obviously,MAGE-A3 may be an important tumor marker for the early diagnosis of cervical disease and the treatment of cervical cancer.MAGE-A3 may be an ideal target for the biological treatment of cervical cancer,which is of great clinical value for further understanding the prognosis of cervical cancer biological behavior.2.The overexpression of MAGE-A9 is closely related to the development of cervical cancer.The overexpression of MAGE-A9 is related to the clinical stage,pathological grade and lymphatic metastasis of cervical cancer.MAGE-A9 may be an early diagnosis of cervical disease,An important tumor marker for cervical cancer treatment.MAGE-A9 may be the ideal target for the biological treatment of cervical cancer.MAGE-A9 has a great clinical value for further understanding the prognosis of cervical cancer biological behavior.
