Objectives To investigate the expression of DNA methyltransferase 1(DNMT1)and Cyclin A1(CCNA1)protein and m RNA in cervical intraepithelial neoplasia and cervical cancer.Methods 90 patients who were diagnosed as LSIL,HSIL and SCC from November 2016 to October 2017 in the Affiliated Hospital of North China University of Science and technology.30 cases whose cervical tissue without such pathological changes were used as control group.Immunohistochemistry was used to investigate the proteins expression of DNMT1 and CCNA1 in cervical tissues.Western blot and qPCR were used to investigate the protein and m RNA expression of DNMT1 and CCNA1 in cervical tissues.MannWhiteny rank sum test was used for grade data,and ?2 test was used for count data.Oneway analysis of variance was used for comparison between groups and SNK test was used for comparison between groups.Results 1 The results from immunohistochemistry told that the expression of DNMT1 protein in HSIL and LSIL were higher than that in control,which were lower than that in SCC.The DNMT1 in HSIL was higher than that in LSIL(P<0.05).The expression of CCNA1 protein in HSIL were lower than that in LSIL and control,which were higher than that in SCC.2 The results of Western blot showed that the positive rates of DNMT1 in SCC,HSIL and LSIL were 73.33%,56.67% and 36.67%,which were higher than that in control(10.00%).The DNMT1 in SCC was higher than that in LSIL(P<0.05).The relative expression of DNMT1 in the HSIL and LSIL were 0.73±0.48 and 0.40±0.29,which were higher than control(0.23±0.10).It were lower than that in SCC(1.37±0.74).The DNMT1 in the HSIL higher than that in LSIL(P<0.05).The positive rate of CCNA1 in SCC(10.00%)was lower than HSIL,LSIL and control(63.33%,93.33% and 100.00%).The CCNA1 in HSIL was lower than that in LSIL(P<0.05).The relative expression of CCNA1 in SCC(0.78±0.53)was lower than HSIL,LSIL and control(1.59±0.55,1.94±0.29,2.05±0.19).The CCNA1 in HSIL was lower than LSIL and control(P<0.05).3 The results from qPCR showed that the positive rates of DNMT1 m RNA in SCC,HSIL and LSIL were 80.00%,63.33% and 43.33%,which were higher than control(16.67%).The DNMT1 m RNA in SCC was higher than that in LSIL(P<0.05).The relative expression of DNMT1 m RNA in the HSIL and LSIL were 1.97±0.79 and 1.13±0.16,which were higher than control(1.00).It were lower than that in SCC(3.14±1.14).The DNMT1 m RNA in HSIL was higher than that in LSIL(P<0.05).The positive expression rate of CCNA1 m RNA in SCC group(6.67%)was lower than HSIL,LSIL and control(60.00%,93.33%,100.00%).The CCNA1 m RNA in HSIL was lower than that in LSIL(P<0.05).The relative expression of CCNA1 m RNA in SCC(0.27±0.22)was lower than HSIL,LSIL and control(0.79±0.27,0.99±0.05,1.00).The CCNA1 m RNA in HSIL was lower than that in LSIL and control(P<0.05).Conclusions With the severity of cervical lesions,the positive expression rate and relative expression of DNMT1 protein and m RNA increased gradually.The positive rate and relative expression of CCNA1 protein and m RNA decreased gradually.The DNMT1 and CCNA1 expression is associated with cervical lesions.
