Tick is a kind of specialized blood-sucking arthropods.More than 800 kinds of ticks have been found around the world and spread over 200 kinds of pathogens like virus,bacteria and protozoa,including 126 kinds of viruses.The major tick-borne viruses are TBEV?CCHFV and SFTSV in China,and reports showed that part of Jilin and Heilongjiang province were epidemic area.Due to the relatively less report of other tick-borne viruses and the undefined background information in northeastern china,it is of great significance to have a research about ticks and tick-borne viruses.Morphological and molecular biology identification of 4318 ticks collected among 8 cities of 2 provinces from March toJune,2016 andhave viral metagenomic studys in samples based on statistical results.3 kinds of viruses which have more contigs due to the viral-metagenmic results are choosed.Ticks were carried out PCR amplification,cloning,sequencing,homology phylogenetic tree analysis and cell isolation in this research?To learn what viruses ticks have in some areas of Jilin and Heilongjiang Province,and to prevent disease propagated cross-border,we start to have a preventive research.Results showed that 4318 ticks of 5 kinds of 3 genuswere collected from 8 cities of 2 provinces from March toJune,2016,We found out I.persuleatuswas the most,D.silvarum was the least.Viral-metagenomic research results showedTBEV,new Nairovirus,and SBV hadhigher contigs.Then we had molecular biology detectionon these 3 viruses,results showed SBV had the highest prevalence about 0.65%,followed by new Nairovirus and TBEV 0.55% and 0.16%.This is the first epidemiological survey on new Nairovirus and SBV around the world.Epidemiological survey also found out that 3 viruses were prevalent in I.persuleatus,new Nairovirus were prevelant in H.loniconistoo,and SBVandnew Nairovirus werealso discovered a little in H.concinna.Besides that,we had the electron microscopy picture of these 3 viruses which showed typical morphological structure of their genus,and it is also the first electron microscopy picture of SBVandnew Nairovirus isolated from tick in the world.
