Objective:To compare the differences between TGF-?1 and PDGF-AB in venous blood,inactivated PRP and diversified activating PRPs from the release curves and the area under the curve in a vitro atmosphere that simulating human body temperature.To provide a clinical evidence of PRP in application rationalization and treatment utilization.Methods:A total of 10 healthy volunteers were selected and the PRP was prepared by Landesberg's method.The platelet concentrations in venous blood and PRP were measured.The activator was made up of bovine thrombin 1000 U and 10% 1ml calcium chloride solution.The Thrombin-Calcium chloride-PRP group was made by PRP and the activator in a ratio of 10:1.The Calcium chloride-PRP group was thus in a ratio of 10:1 by PRP and 10% calcium chloride solution instead.A quantitative sandwich enzyme-linked immunosorbent assays(ELISA)was used to examine the amount of TGF-?1 and PDGF-AB in the Whole blood group?Inactived PRP group?Calcium chloride-PRP group?Thrombin-Calcium chloride-PRP group in 0?1?8?24?72 ? 168 hours.The release curves of TGF-?1 ? PDGF-AB were based on afore-mentioned data,then a comparation of TGF-?1 and PDGF-AB release curves and the areas under the curves in different groups was performed.Results:1.PRP immediately formed into a gel after mixed with thrombin combined and calcium chloride,and the concentration of TGF-?1,PDGF-AB reached the peak in 1h after activation,respectively increased from 41779.14±21009.17pg/ml ?76746.17±17776.39pg/ml to 84339.26±20789.42pg/ml ? 124269.67±34977.13pg/ml,then decreased gradually.The release curve was direct and rapid.The PRP came into a gel state in approximate 1h after mixed with calcium chloride,the concentration of TGF-?1 and PDGF-AB were respectively increased from 11526.87 ± 5024.52 pg / ml?10614.66 ± 4234.61 pg / ml and remained high at 168 h.2.The AUC0-168 h of TGF-?1 and PDGF-AB in the Inactived PRP group were higher than that in the whole blood group(P<0.05),and the AUC0-168 h of TGF-?1 in the Calcium chloride-PRP group was higher than that in the Thrombin-Calcium chloride-PRP group(P<0.05).However,there was no significant difference in the AUC0-168 h of PDGF-AB between the Calcium chloride-PRP group and the Thrombin-calcium chloride-PRP group.Conclusion:1.The platelet rich plasma was prepared by a quadratic centrifugation way to obtain a higher platelet concentration,and the platelet concentration was 7.71 times higher.2.Using calcium chloride as a activator of PRP can get a higher release concentration of TGF-?1?PDGF-AB and a longer release time,with the largest area under the curve.
