Objectives:To compare the differences of serum Apo J level and Apo J gene rs11136000 polymorphism between CHD group and the control group,To investigate the relationship between serum Apo J level and rs11136000 gene polymorphism and CHD,the clinical types and the coronary artery stenosis severity.Methods:We were selected 324 patients with CHD and normal control group from Department of Cardiology in Tianjin Chest Hospital between November 2016 and February 2017.The basic data of the subjects were recorded,including sex,age,smoking history,history of hypertension,history of diabetes,family history of CHD,height,weight,medication situation.On the second day morning after fasting 8 hours were collected fasting venous blood to test lipids,including TC,TG,LDL-c,VLDL-c,HDL-c,Apo A,Apo B and calculating AI.Recorded the results of coronary angiography.Using ELISA method to detect the serum level of Apo J;using DNA extraction kit to extract selected genomic DNA and using PCR-RFLP method to detect the rs11136000 polymorphism of Apo J gene.Grouping methods:(1)according to CAG were divided into CHD group(261 cases),control group(63 cases);(2)according to whether have treatment history of statin divided CHD group into statin group(80 cases)and without use statin group(181 cases);(3)according to the clinical types classification standards divided CHD group into SAP group(36 cases),UAP group(189 cases),and AMI group(36 cases);(4)according to coronary artery stenosis divided CHD group into coronary artery stenosis 50-70% group(52 cases),coronary artery stenosis 70-90% group(155 cases),coronary artery stenosis>90% group(94 cases);(5)according to the genotype of the rs11136000 site,divided into CC group(222 cases),CT group(91 cases),TT group(11 cases).Results:1.There were significant differences in age,gender,smoking history,history of diabetes and hypertension between CHD group and control group(p<0.05),the serum levels of TC,TG,LDL-c,Apo J and AI were significantly higher in CHD group than in control group(p<0.05),while the serum level of HDL-c and Apo A in CHD group was lower than in control group(p<0.05),there were no significant difference in family history,BMI,VLDL-c and Apo B between the two groups(p>0.05);After removal of diabetes,the serum Apo J level in CHD group was significantly higher than that in control group(p<0.05);The serum Apo J levels of CHD patients with DM and DM group were significantly higher than the control group(p<0.05),the serum Apo J levels of CHD patients with DM was higher than the DM group(p<0.05);The levels of serum TC,TG,LDL-c,Apo J,Apo B and AI in stain group were significantly lower than without use statin group(p<0.05).There were no significant difference of HDL-c,VLDL-c,Apo A leves between the two groups(p>0.05).The level of serum Apo J was positively correlated with AI(p<0.05).2.The serum level of Apo J in SAP group and UAP group were significantly higher than the control group(p<0.05),but there were no significant difference between SAP group and UAP group,AMI group(p>0.05),The AI of AMI group,UAP group,SAP group were significantly higher than the control group(p<0.05),the AI of AMI group and UAP group were higher than SAP group(p<0.05),the AI of UAP group is higher than SAP group(p<0.05);The serum Apo J level of Coronary artery stenosis 50-70% group,70-90% group,>90% group,were significantly higher than the control group(p<0.05),but there were no significant difference among the three groups(p>0.05),the AI of three groups were higher than control group(p <0.05),the AI among the three groups were significantly differences(p<0.05),with the aggravation of coronary artery,the level of AI increased(p<0.05).3.The distribution of rs11136000 genotype and allele C,T allele frequency of rs11136000 in Apo J gene had no significance difference betwwen CHD group and control group(p>0.05),and the same as different degree of coronary artery stenosis groups and different clinical types.4.The serum TC,TG,LDL-c,Apo J,Apo B?AI levels of statin group were significantly lower than without use statin group(p<0.05),the HDL-c,VLDL-c and Apo A levels between the two groups showed no significant difference(p>0.05).5.The serum Apo J level of type CC and type CT+TT in patients with CHD were significantly higher than control group(p<0.05),and in CHD group,the serum Apo J level of type CT+TT in statin group was significantly lower than without use statin group(p<0.05).6.Single factor Logistic regression analysis showed that male,age,smoking history,family history of CHD,hypertension,diabetes,TC,TG,LDL-c,Apo J and AI were associated with CHD,HDL-c,Apo A are protective factors for CHD.Multivariate Logistic regressionanalysis showed that the history of diabetes and Apo J were associated with CHD,HDL-c was an independent protective factor for CHD.Conclusion: 1.The serum Apo J level elevated in patients with CHD,atherosclerosis that may cause Apo J compensatory increase,inturn,inhibiting the progression of atherosclerosis,and it may plays an protective effect on CHD;elevated serum Apo J level may serve as a predictor of coronary heart disease.2.The polymorphism of rs11136000 locus in Apo J gene may not be associated with the incidence of CHD,the different clinical types of CHD and the severity of coronary artery lesions.
