Stellera chamaejasme L.is a toxic perennial herb belonging tothe Thymelaeaceae family of flowering plants,and its root is usedin traditional Chinese medicine,also known as Broken grass,was first recorded in "Shennong’s Herbal".It has water expectorant,broken disease insecticide effect.Modern pharmacology and clinical studies have shown that Stellera chamaejasme has anti-HIV,anti-tumor,anti-bacterial,anti-tuberculosis,anti-leukemia,anticonvulsant,antiepileptic and so on.Until now,studies on Langdu are mainly focused on the extraction and separation of certain chemical components,but there are few reports about the qualitative and quantitative analysis of the chemical compositions,processing methods on chemical constituents of Stellera chamaejasme as well as metabolites in vivo.The present study is based on the current research deficiencies:1、Using the MIM-IDA-EPI scanning method,the LTQ / Orbitrap mass analysis method was used to analyze and identify the aqueous extract of Stellera chamaejasme.The results showed that there were 61 compounds(31 flavonoids,14 kinds of coumarins,2 terpenes,6 species of lignans,3 species of phenylpropanone and 13 others)were identified.The possible cleavage pathways were speculated,and the mass spectrometry was analyzed.The method is simple and rapid,which lays a foundation for the comprehensive study on the chemical composition and quality control of Stellera chamaejasme,and provides an effective means for the qualitative identification of Chinese herbal medicines.2、MIM-IDA-EPI detection method using the combination of positive and negative ion mode,the characteristic peak of Stellera chamaejasme before and after processing the application of metabonomics technology to compare the characteristic peaks of Stellera chamaejasme L.,using the method of multivariate statistical method and the combination of pharmacological screening,and according to the information identification of the characteristic peaks of the characteristic peaks.Discovery of Stellera chamaejasme L.and its processed products have obvious differences between the chemical composition and found that after processing through the analysis,there is a significant difference between a peak,which structure 29 different variables have been identified.3 、 To establish a UPLC-MS method for simultaneous determination of neochamaejasmine A,neochamaejasmine B,isochamaejasmine and daphnoretin in rat plasma and to use it in rats after intragastric administration.Pharmacokinetics of four components.The results show that the lower limit of the determination of neochamaejasmine A,neochamaejasmine B,isochamaejasmine and daphnoretin in plasma was 0.274 ng / mL,and the linear range was 0.274 ~ 200 ng / mL(r> 0.99),batch and inter-assay precision(RSD)were less than 10%.The maximum concentration(Cmax)of cephalosporin reached a maximum concentration(Cmax)of 1.48 ng / mL at 0.083 h(tmax)at 0.5 h(tmax)to reach the maximum concentration(Cmax)of 5.24 ng / mL,indicating that the two substances were rapidly absorbed,Rapid access to the blood circulation system.Neonatal toxin B and heterosexins were not exposed in rat plasma.
