Study On The Of Expression Of Phosphodiesterase Type 5(PDE5)in Adenomyosis | | Posted on:2018-01-23 | Degree:Master | Type:Thesis | | Country:China | Candidate:T Wu | Full Text:PDF | | GTID:2334330542467161 | Subject:Obstetrics and gynecology | | Abstract/Summary: | PDF Full Text Request | | Objective: To investigate the expression of phosphodiesterase type 5(PDE 5)in the women with and without adenomyosis;Evaluate the pathogenesis of adenomyosis and provide new ideas for the drug treatment of adenomyosis.Methods: 108 patients were recruited from June 2015 to June 2016 in the Obstetrics and Gynecology of the second affiliated hospital of Soochow university.These patients will undergo hysterectomy for benign diseases.The basic clinical data and etiology were gathered from them.Endometrium and myometrium were collected,according to the results of pathological result,all patients were divided into two groups,namely adenomyosis group with 56 patients and control group with 52 patients.Employed Immunohistochemical streptavidin-biotin peroxidase(SP)method used to detect the expression of PDE 5 in endometrium,myometrium and ectopic endometrium.Compare the expression of PDE 5 in the women with and without adenomyosis in different groups;in different menstrual phase.The relationship between the expression of PDE 5 and dysmenorrhea;the duration of the disease;he degree of tenderness of uterosacral ligaments;the uterine size were analyzed Spearman’s Bivariate Correlation.Results:1、 PDE 5 both expressed in AM group and control group during each menstrual cycle and PDE 5 located in cytoplasm of cells in eutopic,ectopic endometrium and myometrium.2、 The Expression of PDE 5 in eutopic endometrium and myometrium of AM group was significantly stronger than control group(χ2=37.80,P<0.01;χ2=36.62,P<0.01).The Expression of PDE 5 in eutopic endometrium was higher than ectopic endometrium(χ2=12.11,P<0.01).3、 Expression of PDE 5 was no significant difference in proliferative phase compared with secretory phase in two groups(P>0.05).4、 There was no significant difference in expression of PDE 5 between the adenomyosis with or without endometriosis;Expression of PDE 5 was no significant difference in patients with adenomyoma compared with the patients without adenomyoma(P>0.05).5、 There was a positive correlation between the expression of PDE 5 in myometrium of AM group and the degree of dysmenorrhea in patients with adenomyosis(r=-0.71,P<0.05);What’s more,the longer the duration of the disease,the higher the expression of PDE 5 in myometrium(r=0.68,P<0.01);There was also a positive correlation between the expression of PDE 5 in myometrium the degree of tenderness of uterosacral ligaments in patients with adenomyosis(r=0.76,P<0.01).There was no significant positive correlation between the expression of PDE 5 and the uterine size in patients with adenomyosis(P>0.05).Conclusions:1、 PDE 5 both expressed in AM group and control group during each menstrual cycle,and PDE 5 located in cytoplasm of cells in eutopic,ectopic endometrium and myometrium.2、 The expression of PDE 5 in eutopic endometrium and myometrium of AM group was significantly stronger than control group(P<0.01).There was a positive correlation between the expression of PDE 5 in myometrium of AM group and the degree of dysmenorrhea,the degree of tenderness of uterosacral ligaments in patients with adenomyosis(P<0.05).The result suggested that PDE 5 is closely related to the dysmenorrhea in patients with adenomyosis.3、 There was no significant difference expression of PDE 5 between proliferative phase and secretory phase in each group(P>0.05).Which may suggest that there is no correlation between the expression of PDE 5 and the menstrual cycle.4、 The longer the duration of the disease,the higher the expression of PDE 5 in myometrium of adenomyosis patients.It shows that the abnormal expression of PDE 5 is associated with progress of adenomyosis.5、 The expression of PDE 5 is positively correlated with the dysmenorrhea and the progress of adenomyosis.It can offer a new idea that inhibiting the expression of PDE 5 can relieve pain;improve the patient’s life satisifaction and alter the course of the disease. | | Keywords/Search Tags: | Adenomyosis, dysmenorrhea, Uterine contractility, Phosphodiesterase type 5(PDE 5), Cyclic guanosine monophosphate(cGMP) | PDF Full Text Request | Related items |
