Objective: To investigate the effects of berberine,cisplatin and two drugs combined on the proliferation and apoptosis of human lung cancer A549 cells and its possible mechanism.Methods:1?The lung cancer A549 cells were divided into control group,berberine group,cisplatin group and combined group.The control group was fed with equal volume of F12 medium;the berberine group was treated with 10?mol/L ? 25?mol/L ? 50?mol/L ? 100?mol/L berberine;the cisplatin group were added 1mg/L ? 2mg/L ? 4mg/L cisplatin;the combined group were respectively treated with 10?mol/L?25?mol/L?50?mol/L?100?mol/L berberine and 2mg/L cisplatin.The effects of berberine,ciaplatin and two drugs combined on the proliferation of cells were detected by flow cytometry.2?The lung cancer A549 cells were divided into control group,berberine group,cisplatin group and combined group.F12 medium was added into the control group,25?mol/L berberine was added to the berberine group and the cisplatin group was added 2mg/L cisplatin,the combined group treated with 25?mol/L berberine and 2mg/L cisplatin.The effect of berberine,cisplatin and two drugs combined on cell cycle and apoptosis were analyzed by flow cytometry.The effect of berberine,cisplatin and two drugs combined on the expression of Survivin,Bcl-2 and Bax in human lung cancer A549 cells were detected by reverse transcription polymerase chain reaction(RT-RCR)and western-blottiong.Results:1,The inhibitory rate of proliferation of berberine and cisplatin was higer than that of control group(P<0.01).With the increase of berberine and cisplatin concentration,the inhibitory rate of proliferation increased(P<0.01);with the prolongation of the time,the inhibitory rate of proliferation increased(P<0.01).The inhibitory rate of proliferation of the combined group was higher than that of berberine group and cisplatin group,the difference was statistically significant(P<0.01).2,Flow cytometry showed that compared with the control group,the proportion of G1 phase cells in the berberine group increased,the proportion of S phase cells in the cisplatin group increased,and the proportion of G1 phase and S phase cells increased in the combined group(P<0.01).Compard with the control group,the apoptosis rate of the berberine group,cisplatin group and the combined group increased,the difference was statistically significant(P<0.01),The apoptosis rate of the combined group was higer than that of the berberine group and the cispaltin group,the difference was statistically significant(P<0.01),3,The expression of Survivin and Bcl-2 mRNA in the berberine group,cisplatin group and the combined group was lower than that in the control group,the expression of Bax mRNA was higher than that of the control group,the difference was statistically significant(P<0.01);The expression of Survivin and Bcl-2 mRNA in the combined group was lower than that in the berberine group and cisplatin group,the expression of Bax mRNA in the combined group was higher than that in the berberine group and cisplatin group,the difference was statistically significant(P<0.05).4,The expression of Survivin and Bcl-2 protein in the berberine group,cisplatin group and the combined group was lower than that in the control group,the expression of Bax protein was higher than that of the control group,the difference was statistically significant(P<0.05);The expression of Survivin and Bcl-2 protein in the combined group was lower than that in the berberine group and ciaplatin group,the expression of Bax protein in the combined group was higher than that in the berberine group and cisplatin group,the difference was statistically significant(P<0.05).Conclusion:1,Berberine and cisplatin can inhibit the proliferation of human lung cancer A549 cells,that was dose and time dependent.The combination of the two have a better effect on A549 cells.2,Berberine can synergize cisplatin-iduced apoptosis of human lung cancer A549 cells,and its mechanism may be related to up-regualtion of Bax expression,down-regulation of Bcl-2 and Survivin expression.
