| Objective: Objective to investigate the effect of total flavonoids of Portulaca oleracea on chronic prostatitis in mice and rats.In this study,we measured the amount of drinking water,urine output,urine flow dynamics,WBC,Lecithin corpuscle,behavioral index and the content of Ig,IL,TNF,IFN,HIS,BK,COX-2,LTB4,PGE2.This study is to explore the pathogenesis of chronic prostatitis,from the perspective of immune regulation mechanism and oxidative stress response to reveal the therapeutic effect of total flavonoids of Portulaca oleracea on chronic prostatitis and its main mechanism of action in the treatment of chronic prostatitis.Method:The mouse model of chronic prostatitis was established by using the method of removing testis and inducing estrogen.Intragastric administration,Sanjin group,Conprata Pulean group,Large,medium and small doses of total flavonoids of Portulaca oleracea;Estradiol benzoate 0.25mg/kg was injected subcutaneously daily for 30 days.The urine volume and drinking water were respectively measured at Tenth days,twentieth and twenty-ninth days.Determine the light and dark shuttle and cross maze index at twenty-eighth days.After thirtieth days of administration,the blood was collected and the serum was separated.Detected the E2 and T content.After cervical dislocation,the prostate gland,thymus,spleen,kidney,epididymis and bladder were removed.Observed the WBC,Lecithin corpuscle and the content of IL-10,IL-8,TNF-α,and Ig G in the prostate and the morphological changes.Both lateral lobes of the prostate given by 25% Xiaozhiling injection.Eighth days after operation,give medicine by i.g,Sanjin group,Conprata Pulean group,Large,medium and small doses of total flavonoids of Portulaca oleracea.Urine volume and drinking water were measured at tenth and twentieth days.Nineteenth days,conduct rat open field test.Twenty-first days after the operation,the rats were detected by the method of urinary fistula after eighteenth days.Determine of Ig A,Ig G,C3 and CRP in blood serum.Remove prostate gland,thymus,spleen,kidney,testis,epididymis.Observed the WBC,Lecithin corpuscle and the content of IL-10、IL-1β、IFN-γ、TNF-α、HIS、BK、COX-2、LTB4、PGE2 in the prostate and the morphological changes.Result: Chronic prostatitis in mice model builds successfully by testicular castration combined with estrogen induces.In the model group were significantly decreased in the urine volume of drinking water for 20 days,WBC in the prostate tissue significantly increased the density of lecithin corpuscle score decreased significantly(P < 0.01),increase the content of E2 in serum,the content of T was decreased(P < 0.01),IL-8,prostate homogenate TNF-α,Ig G level significantly elevated levels of IL-10 were significantly increased(P < 0.01).Total flavonoids of Portulaca oleracea can be significantly increase in ovariectomized estrogen induced chronic prostatitis caused by prostate tissue in mice model of lecithin corpuscle,significantly reduce the number of WBC(P < 0.01),decrease the content of E2 in serum,increase the content of T in serum(P < 0.05),significantly increase the number of white blood cells and the level of IL-10 in prostate tissue(P < 0.01),and significantly decrease IL-8,prostate homogenate TNF-α,Ig G level(P < 0.01).Improve the mice drinking and urination,increase the amount of water and urine volume of mice,the mice and the cross maze and the shuttle has a certain improvement effect,in addition to Purslane total flavonoids can effectively alleviate the prostate,mice thymus,spleen,epididymis,pathological changes of the bladder.Chronic prostatitis rat model builds successfully by Xiaozhiling injection induced.The model group rats urine quantity and water quantity decreased significantly in 20 days.WBC in the prostate tissue significantly increased the density of lecithin corpuscle score decreased significantly(P < 0.01),serum Ig A,Ig G,C3 and CRP were significantly decreased(P < 0.01),prostate homogenate of IL-1 beta,IFN-gamma,TNF-alpha,HI,BK,COX-2,LTB4 and PGE2 levels were significantly increased,IL-10 level decreaseed significantly(P < 0.01).Obvious pathological changes were found in the pathological tissues.Total flavonoids of Portulaca oleracea can obviously increase the urine volume of twentieth Xiaozhiling induced chronic prostatitis model rats days as water intake(P < 0.01),significantly improve rat urodynamics,reduce rat model of maximum bladder pressure,increase urination interval model rats(P < 0.01),can increase the activity frequency of the model rats in the open the anxiety behavior;significantly reduce the number of WBC in the prostate tissue of model rats,increase the density of lecithin corpuscle in rat model of prostate tissue(P < 0.01),significantly decreaseed in serum of model rats in Ig A,Ig G,C3,CRP,IL-1β and IFN-γ in prostate homogenate of beta,gamma,alpha TNF-α.HIS,BK,COX-2,LTB4 and PGE2 levels and increased IL-10 levels in prostate homogenate(P < 0.01),can effectively improve the rat model of prostate,thymus,spleen,testis,epididymis,pathological changes of the bladder.Conclusion: This study showed that the total flavonoids of Portulaca oleracea can be effectively increased the level of anti-inflammatory cytokine IL-10,thus effectively suppress the inflammatory cytokines IL-1 and IL-8 beta release,thereby reducing Cox-2 and PGE2 levels,thereby reducing the sensory nerve endings of BK,LTB4 and other sensitivity inflammatory substances,relieve the pain reaction of chronic prostatitis animal model,and to reduce the number of WBC in prostate homogenate,increased the density of lecithin corpuscle.Portulaca oleracea total flavonoids can effectively inhibit immunoglobulin release and promote inflammation of C reactive protein and complement 3 metabolism,can improve tissue injury,while the model animal behavior also has improvement effect;the mechanisms mainly through immune regulation,improve systemic and local immune function,to improve the comprehensive therapy effect. |
