Oxidative damage to the body caused by excess free radicals can lead to a variety of diseases.However,there are many toxic and side effects of the applied synthetic antioxidants,and the existing dosage forms of natural antioxidants are not conducive to transportation and storage.Portulaca oleracea L.is a medicinal and food homologous wild plant and has many medicinal values.Some studies have shown that flavonoids abtained from Portulaca oleracea L.have strong antioxidant activity,but research on modern dosage forms of total flavonoids in Portulaca oleracea L.(POTF)as an antioxidant has never been reported.Therefore,this experiment preliminary explored the modern dosage form and antioxidant activity of POTF.In this experiment,POTF,the active components of Portulaca oleracea L.were extracted by ultrasound-assisted alcohol extraction,and the effects of ethanol concentration,extraction time,extraction temperature and solid-liquid ratio on the extraction rate of POTF were investigated by single factor test.Combined with the results of single factor test,the optimum extraction process of POTF by ultrasound-assisted alcohol extraction was determined by orthogonal test.The singlefactor test was used to screen the optimal tablet pressure,filler,binder and excipient formula for POTF tablets.The optimal preparation pressure,filling agent,adhesive and excipient formula of POTF tablets were screened by single factor test.The optimum preparation technology of POTF tablets was determined based on the appearance,weight difference,molding rate,tablet hardness and disintegration time.Further taking vitamin C as a positive control,we measured the scavenging ability of POTF and its tablets,such as nitrogen radicals,hydroxyl radicals,superoxide anion radicals and total antioxidant capacity.The optimal extraction process of POTF with ultrasonic assisted extraction was as follows: the highest extraction rate of POTF was 33.31 mg/g when the ethanol concentration was 50%,the extraction time was 50 minutes,the extraction temperature was 60°C,and the ratio of material to liquid was 1?30.The optimal preparation process of POTF tablets was as follows: the ratio of POTF and excipients was 1?1.5,the formula of excipients was lactose and dextrin(1?1),the adhesive was 4% povidone dry powder,and the production pressure was 7.5 kn.All the obtained tablets were in accordance with the provisions of the Chinese pharmacopoeia.The antioxidant effects of POTF and its tablets was as follows: when the main dosage of POTF,its tablets,and VC reached the highest level,the scavenging rates of nitrogen free radicals were 86.81%,86.93%,and 80.69%.The scavenging rates of hydroxyl free radicals were 94.96%,94.76%,and 98.42%.The scavenging rates of superoxide anion free radicals were 78.78%,78.45%,and 93.59%.The total antioxidant was 69.56 U/mL,70.34 U/mL,and 99.47 U/mL.The antioxidant effects of POTF and its tablets showed that both had strong ability to scavenge nitrogen free radicals and hydroxyl free radicals,and the results were similar to vitamin C.The effects of POTF and its tablets on scavenging superoxide anion free radicals and total antioxidant were different from that of vitamin C.However,there was no significant difference in antioxidant capacity between POTF and its tablets.Therefore,it was concluded that the preparation process and excipients of POTF tablets had no significant effect on the antioxidant activity of POTF.The experimental results provided theoretical basis for POTF tablets as natural antioxidants.
