ObjectiveStudy on the structural changes of green tea polyphenols-functionalized porous calcium phosphate ceramics and their effects on the proliferation and differentiation of bone marrow mesenchymal cells.MethodsWe utilize polyphenol(TP)functionalized porous calcium phosphate ceramics(PCPC).Infrared spectroscopy was used to detect whether PCPC and TP were bound.Simultaneous thermal analyzer was used to detect the thermal stability of PCPC combined with TP.Morphological changes of bone marrow-derived BMSCs were detected by scanning electron microscopy(SEM)and confocal laser scanning microscopy(CLSM).Cell proliferation was examined by MTT analysis.BMP2 expression was analyzed by Western blot and immunofluorescence.Osteocalcin(OCN)and osteopontin(OPN)were analyzed by Western Blot,and osteal mineralization assays were examined using alizarin red staining and ALP activity assay.Finally,MAPK signaling pathways including P-PErk and JNK1 proteins were analyzed by Western Blot and immunofluorescence.ResultsOur results show that tea polyphenols functionalized calcium phosphate ceramic has strong thermal stability,and can make the surface of calcium phosphate ceramic redistributed mineralized nanoclusters.Through a series of studies,we found that PCPC/TP was beneficial to the proliferation and differentiation of BMSCs and up-regulates the expression of BMP2 protein and MAPK signaling pathway-related proteins.Alizarin red staining and ALP analysis showed that PCPC/TP could increase the mineralization ability of BMSCs after osteogenic differentiation.Western Blot analysis of OCN and OPN showed that compared with PCPC,PCPC/TP did not significantly improve the expression of PCPC/TP,but still had good osteoinduction compared with normal condition.ConclusionsThe pore morphology of porous calcium phosphate ceramic after functionalization of tea polyphenol does not change,but it can form remineralized nanoclusters.Otherwise,CPCP / TP relative to CPTP is more conducive to the proliferation and differentiation of BMSCs.
