ObjectiveTo investigate the generation mechanism of radiation induced reproductive system dysfunction of mice by observing the apoptosis of reproductive organs and testing the changes of sex hormone levels in serum,and use the Bushen Jiedu prescription to investigate the protective effect of this drug,explore the protective and therapeutic effect of Bushen Jiedu prescription to the reproductive system damage caused by radiation for further study.MethodsDivided the 72 mice(36 male,36 female)into 6 groups use the random number table method,each group for 12,there were male/female control group,radiation model group,Bushen Jiedu prescription group.Normal eat and drink 2 days for adaptive breeding,Bushen Jiedu prescription group were given 0.2ml Bushen Jiedu granules which the concentration is 1.38g/ml gavage once a day,control group and radiation model group were given 0.2ml normal saline gavage once a day,totally 10 days,on the 11 th day give the radiation model group and Bushen Jiedu prescription group one-time 60 Co ? ray exposure,24 hours after radiation,extract 6 mice in each group use the random number table method to execute,testing the apoptosis of germ cells and the levels of sex hormone;after the radiation,Bushen Jiedu prescription group continue the gavage for 14 days,control group and radiation model group continue gavage the normal saline,normal eat and drink for 14 days,then execute the rest of mice,testing the apoptosis of germ cells and the levels of sex hormone.ResultsObserving the apoptosis of germ cells found that on the 1st day after radiation a small amount of spermatid and sperm in male mice testis were apoptosis,there were little apoptosis cells in Bushen Jiedu prescription group,and the testicular cells are in good condition;on the 14 th day after radiation,there were more apoptosis positive cells in testis of radiation model group,the apoptosis cells were different levels of spermatid in seminiferous tubule,Leyding cell and Sertoli cell,the apoptosis cells also appeared in Bushen Jiedu prescription group,but the number of apoptosis cells were less than radiation model group,and the shape of testis and seminiferous tubule were better than radiation model group,the OD level in radiation model group was higher than control group and Bushen Jiedu prescription group.The ovarian granular cells were apoptosis in female radiation model group on the 1st day after radiation,the shape of oocyte were still normal,the apoptosis granular cells in Bushen Jiedu prescription group were less than radiation model group;on the 14 th day after radiation,the granular cells in ovarian of radiation model group were disorder and apoptosis,and the shape of oocyte were abnormal,at the same time the level of OD was increased,the shape of oocyte in Bushen Jiedu prescription group were almost normal and in good order.Sex hormone levels: male: on the 1st day after radiation,the FSH level in radiation model group were higher than control group(P<0.01);the level of LH in radiation model group and Bushen Jiedu prescription were increased compared with control group(P<0.01);the level of PROG and PRL in radiation model group was significantly higher than control group(P<0.01);for PRL there was only Bushen Jiedu prescription group were higher than control group(P<0.01).On the 14 th day after radiation,the level of FSH,LH,PROG,PRL and E2 was decreased compare with the 1st day after radiation(P<0.05),but it also higher than control group(P<0.05);for the level of T,only the radiation model group was few lower than control group(P<0.05),those index in Bushen Jiedu prescription was very closed to control group(P>0.05).Female: On the 1st day after radiation,the E2 of female in Bushen Jiedu prescription was increased,there was significantly difference compare with the radiation model group(P<0.01);the FSH,PROG in radiation model group was significantly higher than control group(P<0.01);on the 14 th day after radiation,the FSH,PROG in radiation model group was decreased,but it still higher than control group(P>0.05),the E2 in radiation model group and Bushen Jiedu prescription group was lower than control group(P<0.01).ConclusionsIonizing radiation will lead to the abnormal of mice weight,appearance and physiological activity,after the radiation it can test the apoptosis of testis cells in male and ovarian cells in female,but the Bushen Jiedu prescription can effective relieve the appearance of mice,and have positive effects for the situation of reproductive cells apoptosis and the changes of sex hormone level,this shows that the Bushen Jiedu prescription can effective protect the ionizing radiation caused reproductive system damage,it can be the ideal radiation protective agent for further study.
