ObjectiveTo verify the effectiveness and safety of the affinity adsorbent material developed by our research group through HP to remove ETM in dogs.MethodsEndogenous and exogenous ETMmodelswereestablishedby appendectomy and puncture perforation and intravenous injection of ET,the models were divided into the treatment group?n=10?and the control group?n=5?after the success of the modeling,the treatment group was perfused with a hemoperfusion device?DHC-II-340?filled with affinity adsorbent material and the control group was perfused with a blank perfusion apparatus.During the perfusion process,the vital signs were monitored,the levels of endotoxin in the blood,the level of the cytokines and the physiological and biochemical indexes were detected.ResultsEndogenous ETM model was respectively 0.52?1.06?1.95?Eu/ml?in the blood after 6?12 and 24 h,exogenous ETM model was respectively 69.20?105.35?106.01?Eu/ml?in the blood after 30?60 and 120 min;The levels of cytokine TNF-??IL-1??IL-6 and IL-10 were higher than before the model after the endogenous model of 24 h and 120 min.Perfusion therapy:The decrease rate of endogenous and exogenous endotoxin in the treatment group was respectively 99.52%and 94.07%;The cytokine TNF-?,IL-1?,IL-6 and IL-10 in the treatment group were lower than those of the perfusion flow?P<0.05?.The level of endogenous and exogenous PLT in the treatment group was lower?P<0.05?than that at the beginning of perfusion?219.43±8.38 vs 191.71±5.47???222.50±14.19 vs 189.71±15.57?;In the treatment group,liver function?ALT?AST?ALP?TP?GLO?and renal function?Urea?Cr?UA?were decreased after treatment with endogenous and exogenous perfusion compared with the beginning of perfusion?P<0.05?;The electrolyte(K+?Na+?Cl-?Ca2+?P3-?Mg2+?ECO2)had no obvious fluctuation before and after perfusion.ConclusionsThe endogenous and exogenous ETM model was successfully established by used the method of appendectomy and perforation and direct intravenous injection of ET,which laid the foundation for the follow-up study.The affinity adsorbent material developed by our research group can efficiently remove endotoxin and elevated cytokines in dog's blood by perfusion.The vital signs and physical and chemical indicators in the experiment are relatively stable,reflecting the safety of the experiment.
