Objective:To investigate the association between of the single nucleotide polymorphisms(SNPs)rs3764650,rs3752246,rs4147929,rs3752242 of ABCA7 gene with sporadic Alzheimer's disease(SAD)of Han and Kazak nationality in Xinjiang Province.Methods:Genotypes were determined by Kompetitive Allele Specific PCR(KASP PCR)technology in 118 Xinjiang Han and Kazak people SAD patients and 139 healthy controls.The association between the polymorphism and AD was estimated with the odds ratios(OR).Results:Three genotypes of TT,TG and GG were found in the rs3764650 locus of ABCA7 gene.There was no significant difference between the SAD group and the non-AD group(?~2=5.209,P=0.074>0.05).The T and G alleles were between the two groups.There was a significant difference in overall distribution(?~2=4.876,P=0.027<0.05).The frequency of T allele was significantly higher than that of the control group,which may increase the risk of SAD(OR=1.636,95%CI 1.054-2.537).The frequency of G allele was significantly lower than that of the control group,which may reduce the risk of SAD(OR=0.611,95%CI 0.394-0.948).Compared with the control group,the overall difference of the three genotypes in the SAD group ABCA7 gene rs3764650 in Xinjiang Kazak people was statistically significant(?~2=10.9,P=0.004),the frequency of TT genotypes in the SAD group was higher than that in the control group(77.6%>51.9%,?~2=9.444,P=0.002),the frequency of T allele was significantly higher than that in the control group(88.8%>72.8%,?~2=10.542,P=0.01),it may increase the risk of SAD(OR=2.96,95%CI 1.509-5.818);the frequency of G allele was significantly lower than that in the control group(11.2%<27.2%,?~2=10.542,P=0.01),it may reduce the risk of SAD(OR=0.338,95%CI 0.172-0.663);There was no significant difference in genotype and allele frequencies between the AD Han and non-AD groups(?~2=0.037,P=0.982>0.05;?~2=0.025,P=0.875>0.05);Kazak and Han AD groups In comparison,there was no significant difference in the genotype frequency distribution of rs3764650(?~2=4.874,P=0.056>0.05);however,there was a significant difference in the frequency distribution of T and G alleles(?~2=5.934,P=0.022<0.05;OR=2.404),95%CI1.171-4.938).There was no statistically significant difference between the three genotypes and alleles of the ABCA7 gene rs3752242,rs3752246 and rs4147929(P>0.05).Conclusion:ABCA7 gene rs3764650 are associated with the onset of Kazak in xinjiang people SAD,including TT genotype,T allele may be SAD protection factors of the disease,its smallest allele G low expression level is associated with higher risk of AD,there may be racial differences in the incidence of ADrs3764650 sites polymorphism.ABCA7gene rs3752246,rs3752242,rs4147929 loci polymorphisms may have nothing to do with the occurrence of sporadic AD in Han and Kazak people of Xinjiang.
