Objective:To study the effect of turmeric volatile oil(TVO)on the proliferation,migration and invasion of human skin squamous cell carcinoma cell line SCL-1 in vitro,and to explore the mechanism of its apoptosis.Methods:The effects of different concentrations and time(24h?48h?72h)on the proliferation of SCL-1 cells were detected by MTT.The effects of TVO on migration and invasion of skin squamous cell carcinoma SCL-1 cells were examined by Wound scratch assay and Transwell.Flow cytometry analysis and microscope to determine the apoptosis rate of SCL-1 cells induced by TVO.Western Blot detects Survivin and Caspase-3 protein expression.Results:MTT Cell scratch assay and Cell invasion assay suggested that TVO proliferation,migration and invasion of SCL-1 cells in vitro has obvious inhibitory effect(P<0.05),MTT assay showed that TVO can effectively inhibit the proliferation of skin squamous cell carcinoma SCl-1 cells increased with the concentration of TVO and TVO on the long time,SCL-1 cell proliferation inhibiting effect more obvious.In a certain time and concentration dependent manner.Observation under inverted microscope by TVO(5? 10? 20?40? 80mg/L)in SCL-1 cells treated with 48 h,compared with control group,The SCL-1 cell proliferation rate slow down,the wall is gradually reduced,with the higher drug concentration cell gap increases more obvious;visible cells became small,round,and even off,visible floating cells.results showed that treatment with TVO of SCL-1 cells by flow cytometry,with TVO concentration increasing,the apoptosis rate was significantly increased,80mg/L TVO inhibition rate was 33%(P<0.05).Choose a relatively low concentration of TVO(10?20mg/L)for the final concentration of the experimental group by cell scratch test and cell invasion test to detect TVO of skin squamous cell carcinoma SCL-1 cell migration and invasion effects,results,and the control group(DMSO group)compared to TVO significantly reduced the migration and invasion of skin squamous cell carcinoma(P<0.05).Western blot:TVO detection of SCL-1 cells after 48 h,the expression Caspase-3 protein concentration was higher than that of the control group,Survivin protein expression was lower than the control group,the expression of 20mg/LTVO in the biggest changes.The results showed that TVO could significantly increase the expression of apoptosis related protein Caspase-3(P<0.05)and down regulate the expression of Survivin protein.Conclusion:Weastern Blot test showed that the expression of Caspase-3 protein in TVO cells after SCL-1 treatment was higher than that in the control group.The expression of Survivin protein was lower than that in the control group.The results suggest that TVO can promote the expression of apoptosis related protein Caspase-3 and Survivin protein.caspase-3 protein expression increased gradually and significantly higher than the control group,the concentration of TVO was positively related with the amount of protein;and compared with the negative control group,the experimental group can significantly reduce the expression of survivin protein,and negatively related to the concentration of TVO can increase,the expression of Caspase-3,enhance that TVO induce apoptosis.
