Objective LBP can protect the optic nerve cells from ischemia reperfusion injury,but the effect of LBP on cerebral ischemia reperfusion injury in diabetes is not clear.The purpose of this study is to investigate whether LBP can reduce the injury of blood brain barrier related factors in vascular endothelial cells under high glucose and hypoxia conditions.Methods In this study human umbilical vein endothelial cell(HUVEC)was cultured in vitro.We use high glucose and hypoxia to simulate diabetic cerebral ischemia-reperfusion injury,and then observe the effect of LBP on vascular endothelial cells under high glucose and hypoxia.First,We investigate the injury about blood brain barrier related factors reduced by high glucose and hypoxia on HUVEC cells.HUVEC cells was divided into 8 groups :normal glucose group(NG control),high glucose group(HG control),normal glucose hypoxia6 h group(NG+OGD6h),high glucose hypoxia 6h group(HG+OGD6h),normal glucose hypoxia 6h complex oxygen 6h group(NG+OGD6h/R6h),high glucose hypoxia 6h complex oxygen 6h group(HG+OGD6h/R6h),normal glucose hypoxia 6h complex oxygen 24 h group(NG+OGD6h/R24h),high glucose hypoxia 6h complex oxygen 24 h group(HG+OGD6h/R24h);After OGD6h/R6 h,study observes the changes of morphology,proliferation and expression of ZO-1,Occludin and MMP-9 in HUVEC cell were detected in LBP group.HUVEC cells was divided into 6 groups: normal glucose group(NG control),high glucose group(HG control),normal glucose hypoxia 6h complex oxygen 6h group(NG+OGD6h/R6h),high glucose hypoxia 6h complex oxygen 6h group(HG+OGD6h/R6h),normal glucose hypoxia 6h complex oxygen 6h groupwith LBP intervention group(LBP+NG+OGD6h/R6h)and high glucose hypoxia 6h complex oxygen 6h group with LBP intervention group(LBP+HG+OGD6h/R6h).Results When the glucose concentration was 50 mmol\L,the cell activity was 72.33%+ 6.84% compared with the control group;Compared with the control group,the cell activity of the NG group and the HG group were decreased at OGD6h;Compared with the NG group,the cells in the HG group were crinkled and the cell spacing increased;Cell shrinkage increases in group NG and group HG at OGD6h;Cell shrinkage increases markedly in group NG and group HG at OGD6h/6h;The injury of cell morphologic were reduced,but compared with the control group,the cell has little shrinkable;Before hypoxia,compared with NG group,there is Low level expression of ZO-1 and Occludin and hjgh level expression of MMP-9 in HG group;Compared with the control group,The expression of ZO-1 and Occludin decreased in group NG and HG group atOGD6 h and OGD6h/R6 h,and reached the lowest at OGD6h/R6 h,The expression of MMP-9 increased in group NG and HG group and reached the highest at OGD6h;At OGD6h/R6 h,compared with HG group,the cell activity of the LBP+HG group were increased,the degree of cell shrinkage was decreased,the expression of ZO-1 and Occludin was increased,the expression of MMP-9 was decreased.Conclusion 1.High glucose and hypoxia may reduce the expression of Occludin and ZO-1 by increasing the activity of MMP-9 and cause the injury of HUVEC cells2.Under the condition of high glucose and hypoxia,LBP can increase the expression of Occludin and ZO-1 by reducing the activity of MMP-9 and reduce the injury of HUVEC cells.
