Objectives To observe the ability of Lycium barbarum polysaccharides?LBP?to repair traumatic optic neuropathy?TON?in rats and to explore the possible mechanism of Lycium barbarum polysaccharides to traumatic optic neuropathy.Methods 96 healthy and male SD rats were randomly divided into control group,model group,treatment groups of different concentrations of LBP[100,200,400 mg·?kg·d?-1]?A?B?C?and prevention groups of different concentrations of LBP[100,200,400mg·?kg·d?-1]?I?II?III?,each group has 12 rats.In addition to the control group,the remaining 7 groups were treated with 50N optic nerve clip in the left eye on optic nerve injury model.The rats in the control group and the model group were treated with normal saline for 3 months.TheA?B?C group were given daily corresponding doses of LBP[100,200,400mg·?kg·d?-1]for 3 months;The I?II?III group were given corresponding doses of LBP for 14 days before injury,then they were treated with corresponding doses of LBP sequentially for 3 months.All animals were taken with retinal specimens and optic nerve after 3 months.The ultrastructural changes of the optic nerve were observed under electron microscope,and the retinal optic ganglion cells?RGCs?inn the unit area were counted by fluorescence gold labeling.In additon,the activities of superoxide dismutase?SOD?and malondialdehyde?MDA?in the retina need to be measured.Results The optic nerve has changed in the injured group,including axon disappearance and myelin degeneration.Compared with the control group,optic nerve axons?myelin sheath and other morphology have changed in LBP treatment group andprevention group,however,which were significantly lighter than those in injury group.The ability of the optic nerve to repair in high doses was higher than those in the low dose.The number of RGCs were?1103±100?,?1247±36?,?1409±51?and?1185±84?,?1329±50?,?1492±45?inA?B?C group and I?II?III group,which were significantly higher than those in the injury group?802±82?,the difference was statistically significant?P<0.05?.The number of RGCs increased with the increase of LBP concentration in a certain range.The differences of SOD content and MDA activity between the eight groups were statistically significant?P<0.05?.The levels of SOD in A?B?C group and I?II?III group were?38.5±9.22???50.59±7.39???64.97±10.37?and?41.15±9.64???52.26±7.58???66.53±10.35?[U·?mgprot?-1],which were lower than those of the control group?78.26±9.43?[U·?mgprot?-1]?P<0.05?,at the same time,which were higher than those of the model group?23.99±7.55??P<0.05?.The content of MDAin A?B?C group and I?II?III group were?38.42±7.09???24.06±2.94???14.24±3.51?and?32.68±8.75???23.76±6.48???15.21±4.55?,which were higher than those of the control group(?5.1±1.94??P<0.05?,nevertheless,which were lower than those of the model group?52.41±13.34??P<0.05?.In a certain range,the contents of SOD and MDA were also dose-dependent on LBP concentration,the contents of SOD increased with the increase of LBP concentration whereas the contents of MDA decreased with the increase of LBPconcentration.Conclusion 1.Lycium barbarum polysaccharides has protective effect on traumatic optic neuropathy in rats,including improving the ultrastructural changes ofoptic nerve,slowing down or inhibiting the progressive decline of retinal ganglion cells.2.The protective effect of lycium barbarum polysaccharides on TON in rats may be related to up-regulate SOD expression,to inhibite MDA production,and to reduce damage to the optic nerve of reactive oxygen free radical?ROS?,which depends on LBPconcentration in a certain range.
