Objective To screen out antituberculotic loaded bone cement with the best physical properties,mechanical strength and sustained release properties through this study.Methods Preparation of antituberculotic loaded bone cement standard specimens according to the ISO 5833:2002 standard.Ten anti tuberculosis drugs pyrazinamide,isoniazid,rifapentine,protionamide,capreomycin,rifampicin,moxifloxacin,amikacin,ethambutol and streptomycin were mixed with four kinds of non antibiotic bone cements Palacos R,Palacos MV,Simplex P and CEMEX XL in the same proportion of 1.5g:40g and 2.5g:40g separately in the experimental group.In the control group,four kinds of bone cements were mixed with their liquid monomer in the same method.Determine the following performance:(1)physical properties(average dough time,average solidification time and average maximum temperature),(2)extrusion,bending and hard material indentation,(3)mechanical strength(compression strength,bending strength and flexural modulus),(4)Drug concentration in ten time point extract of 1d,3d,7d,14 d,30d,45 d,60d,90 d,120d and 150 d.Results 1.5g and 2.5g group of RFP,INH,RFT and PTO were mixed with four bone cements,the average dough and coagulation time were longer than those of the control group and ISO 5833 standard.The average maximum temperature was in line with the ISO 5833 standard,but it was significantly lower than that of the control group.Then mixed with Palacos R,Palacos MV and Simplex P,each group of RFP,INH,RFT and PTO harden separately in 30 d,14d,8d and 12 d.Then mixed with CEMEX XL bone cement,each group of RFP,INH,RFT and PTO harden separately in 35 d,18d,10 d and 14 d,the compressive strength,bending strength and flexural modulus of the hardening were all < ISO 5833 standard.1.5g and 2.5g group of EB,PZA,SM,CM,AM,MFX mixed with Palacos R,Palacos MV,Simplex P bone cements,the average dough and average solidification time,the average maximum temperature,the compressive strength,the bending strength and the flexural modulus were all in accordance with the ISO 5833 standard,and 20 min hardening after mixing.Ten drugs were mixed with CEMEX XL bone cement,the average dough and solidification time > ISO 5833 standard,the mechanical strength decreased obviously,the hardening time was significantly prolonged,and the control group was also the same.Group of PZA 1.5g and 2.5g,INH 1.5g and 2.5g,RFT 1.5g and 2.5g,PTO 1.5g and 2.5g,were mixed separately with Palacos R,Palacos MV and Simplex P bone cements,the duration of release of drug release in PBS buffer solution can sustainable contract about 45 d,60d,60 d,45d,60 d,45d,150 d,150d,150 d,150d,45 d,60d,90 d,90d,60 d and 90 d.Only when the physical properties,mechanical strength and slow release properties of the drug loaded bone cement conform to the ISO 5833 standard,can they be used for the preparation of drug loaded bone cement.Therefore,PZA,CM,MFX and AM can be mixed with Palacos R,Palacos MV,and Simplex P bone cements,which can be used for the preparation of drug loaded bone cement.Conclusion 1.5g and 2.5g group of PZA,CM,MFX and AM were mixed with Palacos R,Palacos MV and Simplex P bone cements,can be used for the preparation of drug loaded bone cement.1.5g and 2.5g group of RFP,INH,RFT and PTO mixed respectively with Palacos R,Palacos MV,Simplex P and CEMEX XL bone cements,they were all not suitable for the preparation of anti tuberculosis drug bone cement,CEMEX XL bone cement was not suitable for drug loading.
