Objective:Echinococcosis is a serious zoonotic parasitic disease in the pastoral area of western China.This disease which is also known as hydatid disease is caused by the infection of echinococcosis(Echinococcus granulosus).Studies have shown that IL-21 and IL-22 are effective to the pathogenesis,development and treatment of parasites and tumors.Enzyme linked immunosorbent assay(ELISA)was used to detect the serum levels of IL-21 and IL-22 in patients' serum and analyze the correlation with serum clinical indicators such as WBC,ALT,TBIL in patients,in order to find some clinical significance.Methods: 41 cases(22 females and 19 males)of the Tibetan patients with HAE were collected between March and September 2017 from the Department of the Hepatobiliary Surgery,the Affiliated Hospital of Qinghai University.Meanwhile we chose 40 the Tibetan healthy volunteers(17 females and 23 males)as the healthy control group.We did tests blood routine,live function,blood coagulation parameters,immunoglobulin G(Ig G)targeting echinococcus,abdomen Routine Enhanced Scan and so on,detected IL-21 and IL-22 levels in serum by ELASA method on these samples.We compared and analyzed the levels and differences of IL-21 and IL-22 between these two groups,and then tried to find the pertinence of clinical indexes of patients of HAE.Results:The levels of IL-21 and IL-22 in peripheral blood were higher in the HAE group than in the control group,the difference has Statistical significance(P<0.05).Patients with HAE were divided into liver function A,B,C groups according to their liver function,each group has to compare with normal group.The serum levels of IL-21 and IL-22 of the liver function C group were significantly higher than those within liver function A,B and normal groups(P<0.05).Serum levels of IL-21 and IL-22 of liver function A and B groups were significantly higher than normal group(P<0.05).HAE patients are divided into positive group,weak positive group and negative group according to the result of immunoglobulin G(Ig G)targeting Echinococcus,each group has to compare with the normal group.IL-21,IL-22 in hepatic hydatid Ig G positive,weak positive,negative group are higher than normal subjects,the difference has Statistical significance(P<0.05).The serum levels of IL-21 and IL-22 in patients with HAE were analysed correlation with leukocyte,erythrocyte,platelet,total bilirubin,direct bilirubin,indirect bilirubin,alanine aminotransferase,aspartate aminotransferase,activated partial thromboplastin time and focal size.There was a positive correlation between IL-21 and direct bilirubin and total bilirubin,(P<0.05).There was a positive correlation between IL-22 and direct bilirubin,(P<0.05).No evidence shows such correlation with others.Conclution:IL-21 and IL-22 are highly expressed in HAE,and IL-21 is associated with total bilirubin and direct bilirubin.IL-22 is associated with direct bilirubin,suggesting IL-21,IL-22 may be involved in HAE-induced immune response and the occurrence and pathogenesis,and to some extent reflect the liver function and biliary tract injury in patients with HAE.The specific mechanism needs further study.
