Objective To study the antioxidant damage and apoptosis of Lycium ruthenicu m Procyanidins on the cervical dorsal skin of mice with D-galactose subcutane ous injections combined with UV radiation(UVA+UVB).Methods 50 kunming femal mice with Specific pathogen Free(SPF)were divided into 5 groups,blank control group,model control group,positive control group,high dose group,low dose group.Blank control group was normally fed,the other groups were given subcutaneous injection of 5% D-galactose(10ml/kg)and were irradiated UV(UVA+UVB).From the 11 th day,blank control group and model control group were given saline,high dose group and low dose group were given procyanidins at 50mg/kg/ and100mg/kg/d espectively,Vitamin E to positive control group,all the five groups were periodicity treatment for 30 days.Changes of the organization structure and Subcutaneous blood vessels of mice skin were observed content variation of ROS ?CAT activity was tested by Enzymes and biochemical method.Bcl-2?Bax and IL-1?IL-6?TNF-? levels in blood serum were detected by ELISA.Results(1)The model control group appeared photoaging such darken colour,coarse,deep wrinkles and scale compared to normal group,the skin colour was significantly improved and the wrinkles and scales decreased compared with the model group(P<0.05);(2)In the model group,the subcutaneous blood vessel was angiectatic and dark red with nodules and petechiae.The angiectatic blood vesse was reduced and the blood stasis was less after the treatment of VE and procyanidins.The blood vessels in the procyanidins were mildly hyperimic with few nodules,the subcutaneous vessels improved significantly with the increase of the dose while vascular dilatation of VE does not change obviously compared to procyanidins,the difference was statistically significant(P<0.05);(3)The content of ROS,Bax increase significantly and the activity of CAT,the content of Bcl-2 decreased in skin tissue and the levels of inflammatory factor in blood serum increased of model group compared to normal group;the difference was statistically significant(P<0.05);The content of ROS,Bax decreased and the activity of CAT,the content of Bcl-2 increased in skin tissue and the levels of inflammatory factor in blood serum decreased significantly after the treatment of VE and Procyanidins,the differences were statistically significant(P<0.05);(4)ROS,Bax decreased and CAT,Bcl-2 increased in skin tissue and the levels of inflammatory factor in blood serum decreased obviously after the treatment of Procyanidins compared to VE,the difference was statistically significant(P<0.05).(5)ROS,Bax decreased and CAT,Bcl-2 increased in skin tissue distinctly and the levels of inflammatory factor in blood serum decreased significantly with the increase of dosing concentration,the difference was statistically significant(P<0.05).Conclusion Possibly,it has the ability of scavenging free radicals by reducing the content of active oxygen(ROS),enhancing thie vitality of catalase(CAT),on the other hand,it can inhibit the Bax activation and promote the expression of Bcl-2,inhibit cell apoptosis,further reducing the content of inflammatory cytokines to light aging skin protection.
