Objective: This paper aims to explore the influence of Acer truncatum oil damage in skeletal muscle of rats after exhaustive exercise,for Acer truncatum oil as a novel antioxidant and services provide for sport.Methods: The Acer truncatum kernel peeling,purifying air circulation drying oven fully charged hydraulic press to squeeze oil Acer truncatum.Supercritical CO2 technology Acer truncatum continuous oil extraction,set the single factor experiment,control the extraction pressure,temperature and CO2 flow measured after extraction efficiency,optimize the extraction process through the response surface.Selection 7-week-old male SD rats,weighing 270 ± 30 g,after acclimatization week,they were randomly divided into quiet group?CG?,exhaustive exercise group?EG?,low-dose Acer truncatum oil exhaustive exercise group?LG?,middle-dose Acer truncatum oil exhaustive exercise group?MG?,high-dose Acer truncatum oil exhaustive exercise group?HG?,n=8,natural light,free access to water consumption,ambient temperature at 25±2?,country standard molars animal feeding,bedding replaced every 2-3 days.Exhausting rats at nine o'clock every day to running at 15m/min speed for 3 days 15-20 min for adaptive training,after adaptive training the end of the establishment of Incremental Exhausting model for a period of six weeks,and the last training to exhaustion with ether anesthetized rats in the control group and exhaustive groups,abdominal aortic blood on ice to quickly remove the leg gastrocnemius,clipping 0.5cm long and wide organization into fixative prepared tissue sections,the rest of the gastrocnemius after the liquid nitrogen 30 min moved-80? refrigerator spare.Preparation of serum,determination the CK,LDH activity;determination MDA and GSH's content,CAT,SOD,GSH-PX,T-AOC activity and Caspase-8,Caspase-3 activity after gastrocnemius muscle homogenates prepared.Result:1,Through the single factor experiment,confirmed the continuous extraction of supercritical CO2 fluid in the process of the acer truncatum oil,extracting pressure?Mpa?,temperature???,CO2 flow rate?L/h?on the extraction efficiency of influence law;by extracting pressure?Mpa?,temperature???,CO2 flow rate?L/h?as the independent variable,based on extraction efficiency response index,using the Design Expert7.1.3 software Box-Bebnken Design optimization experiment,determine the best process conditions for:28 Mpa extraction pressure,extraction temperature 37.66?,CO2 flow rate of 267.67 L/h,the extraction rate of 96.23%.The fatty acid composition and percentage of acer truncatum:0.15% palmitoleic acid?C16H30O2?,4.13% palmitic acid?C16H32O2?,1.76% ?-linolenic acid?C18H30O2?,35.86% linoleic acid?C18H32O2?,26.53% oleic acid?C18H34O2?,2.6% Stearic acid?C18H32O2?,8.36%11-20 carbon olefine acid?C20H38O2?,0.22% arachidic acid?C20H40O2?,and 13.01% erucic acid?C22H42O2?,0.89% behenic acid?C22H44O2?,4.98% nervonic acid?C24H46O2?,0.34% lifnoceric acid?C24H48O2?,unsaturated fatty acid content of 93.25%.2,Exhaustion exercise instantly,EG group of skeletal muscle disorder and muscle cell's nuclear distribution uneven;Compared with CG group,EG group of serum CK and LDH activity increased;Compared with CG group,EG group of skeletal muscle of MDA,SOD,CAT and GSH-PX,T-AOC,Caspase-8 and Caspase-3,GSH levels drop.3,Acer truncatum oil intervention after exhaustion exercise rats,LG skeletal muscle injury is smaller than EG group,but HG skeletal muscle damage serious than EG group;Compared with the CG group,LG,MG,CK,LDH activity HG group were increased;Compared with the EG group,LG group of CK,LDH activity;MG,HG group CK activity increased than EG group,but lower in the EG group in the LDH activity;Compared with EG and CG group,LG group of MDH content decreased,but the GSH,SOD,CAT and GSH-PX,T-AOC rise;Compared with EG and CG group,the MDA content of MG and HG group to rise,but GSH,SOD,CAT and GSH-PX,T-AOC reduced.Compared to the EG group and LG group,MG and HG Caspase-8,Caspase-3 activity decreases;And LG and CG group of Caspase-8,Caspase-3 activity similar result,compared to the EG group Caspase-8 and Caspase-3 activity of MG and HG group are reduced.Conclusion:1.The corresponding surface optimization CO2 supercritical extraction can improve the efficiency of acer truncatum oil extraction;extraction efficiency is as high as 96.23%,unsaturated acid content as high as 93.25%.2,Low doses of acer truncatum oil can obviously alleviate the skeletal muscle of exhaustion exercise leads to oxidative stress damage.It performance in order to improve the burnout of rat skeletal muscle fibers arranged,muscle nuclear distribution,reduce exhaustion exercise rats serum CK and LDH activity,reduce the skeletal muscle MDA content,improve the activity of antioxidant enzymes,and decrease skeletal muscle Caspase-8 and the activity of Caspase-3.
