Objective: To investigate the effect of mRNA expression and protein expression on Chemerin in visceral adipose tissue,plasma Chemerin concentration and insulin resistance of elderly obese rats were discussed through the intervention of resveratrol combined with exercise in 8 weeks.Methods: Selecting 6 aged rats as the blank control group,another 36 elderly obese rats were randomly divided to 6 groups,6 rats per group,respectively,for the obese control group,static complementary Res group,low intensity exercise group,Res combined with low intensity exercise group,moderate intensity exercise group,Res combined with moderate intensity exercise group.The running speed of the low and medium intensity movement were respectively 12m/min and 15m/min,15min/time,4groups,rest between groups 5 min,1h/d.Using 52.5mg/kg of Res to fill the stomach,5times/w,8 weeks of continuous intervention.8 weeks later,using qRT-PCR technology measuring mRNA expression on Chemerin in visceral adipose tissue,using Western Blot technology measured protein expression on Chemerin in visceral adipose tissue,using ELISA technology measured plasma Chemerin concentration and insulin resistance,determination of blood glucose by automatic biochemical analyzer,calculating insulin resistance.Results:1.Effect of Res combined with exercise on the mRNA expression on Chemerin in visceral adipose tissueThere was a slight increase in mRNA expression on Chemerin in visceral adipose tissue of aged obese rats,but there was no significant difference(P>0.05),low and moderate intensity exercise had little effect on its expression(P>0.05),simply supplemented Res could increase its expression(P< 0.05),Res combined with exercise had little effect on its expression(P>0.05).2.Effect of Res combined with exercise on the protein on Chemerin in visceral adipose tissueThe protein expression on Chemerin in visceral adipose tissue of obese rats was significantly decreased(P<0.01),low and moderate intensity exercise significantlyincreased the protein expression(P<0.01),moderate intensity exercise was higher than low intensity exercise(P<0.01).Simply supplemented Res significantly reduced its expression of protein(P<0.01),Res combined with exercise significantly reduced its protein expression(P<0.01).Res combined with moderate intensity exercise was significantly higher than Res combined with low intensity exercise(P<0.01).3.Effect of Res combined with exercise on the blood glucose and HOMA-IR and plasma Chemerin concentrationThe blood glucose and HOMA-IR were significantly increased in the aged obese rats(P<0.01),Chemerin concentration had no obvious change(P>0.05).Low and moderate intensity exercise could significantly reduce blood glucose,HOMA-IR(P<0.05,P<0.01),less affected by exercise intensity,but it had little influence on Chemerin concentration(P>0.05).Simply supplemented Res had little effect on blood glucose and HOMA-IR(P>0.05),Res combined with exercise could significantly reduce blood glucose and HOMA-IR(P<0.05),but it had little influence on Chemerin concentration(P>0.05).Res combined with exercise could significantly reduce blood glucose(P<0.05).Res combined with low and moderate intensity exercise could reduce HOMA-IR(P<0.05).Conclusion:The following conclusions were drawn from this study.(1)Effect of Res combined with exercise on mRNA expression on Chemerin in visceral adipose tissue of aged obese rats had no significant changes,exercise and exercise intensity had little effect on it,simply supplemented Res could increase its expression,Res combined with exercise had little effect on its expression.(2)The protein expression on Chemerin in visceral adipose tissue of aged obese rats was significantly decreased,exercise could increase its expression,affected by the intensity of exercise was more obvious,simply supplemented Res and Res combined with exercise could reduce its expression of protein,simply supplemented Res more significantly decrease.(3)The blood glucose and HOMA-IR were significantly increased in the aged obese rats.Plasma Chemerin concentration had no obvious change,Res combined with exercise had no obvious effect on plasma Chemerin concentration,but could significantly reduce blood glucose and HOMA-IR.
