JAK2/STAT3 signaling pathway is an important pathway to regulate cell proliferation,differentiation and apoptosis,and is involved in the signal transduction mediated by various cytokines and growth factors.It has been found that JAK2/STAT3 signaling pathway may affect the development of skeletal muscle by regulating the expression of skeletal muscle development related genes.Aerobic exercise can significantly promote the growth of skeletal muscle fibers in developing rats.Resistance exercise can increase the mass and volume of skeletal muscle cells in the recovery period after exercise.Therefore,this study aims to explore the role of JAK2/STAT3 signaling pathway in the skeletal muscle growth of SD rats,and explore the effect of aerobic and resistance exercise on skeletal muscle JAK2/STAT3 signal pathway and the growth and development.This will provide a theoretical basis and scientific basis for promoting the growth and development of skeletal muscle.Purpose:Based on the SD rats 6 weeks of aerobic and resistance exercise intervention,to detect the expression of JAK2/STAT3 signaling pathway and MyoD,Myf5,and investigate the effect of expression level of aerobic exercise and resistance exercise on JAK2/STAT3 signaling pathway and MyoD,Myf5,and promote skeletal muscle growth,and provide theoretical basis for promoting muscle growth and developmentMethods:4 week old male SD rats,with a body weight of 178.66 ± 1.69(g).Randomly selected 24 rats were divided into the control group(C),aerobic exercise group(A),the resistance exercise group(R),each group of 8 rats,fed with standard feed.Aerobic exercise group rats to after training at the speed of 9 m/min 30min in treadmill training,training 5 days a week,4 weeks of training;the resistance training group after training to adapt to increasing weight of the load in the ladder training 3 days a week,4 weeks of training.Incremental load are 40%body,60%body weight,80%body weight,100%body weight,120%body weight,140%body weight,160%body weight(to the end).3/group,the 3 group/day,each time to rest for 30 seconds-60 seconds,each group rest 120-180 seconds.Training 3 days a week,training for 4 weeks.The content of IL-6 in serum ELISA detection;RT-PCR detection of the expression of IL-6,IL-6R,JAK2,STAT3,MyoD,Myf5 mRNA in quadriceps femoris;Western Blotting expression detection of IL-6,JAK2,p-JAK2,STAT3,p-STAT3,MyoD,Myf5 protein in quadriceps femoris.Results:(1)The expression of JAK2 mRNA in the femoral head of rats was significantly decreased after the aerobic exercise for 6 weeks,the expression of STAT3 mRNA was significantly increased,and the content of MyoD and Myf5 protein was significantly increased.(2)The body weight of rats increased significantly after 6 weeks resistance exercise,unit four biceps/body weight ratio increased significantly,the expression of mRNA and IL-6R stock four my-JAK2 mRNA,STAT3 mRNA,MyoD mRNA,Myf5 mRNA,p-JAK2,JAK2 increased significantly,STAT3,p-STAT3,MyoD,Myf5 protein content was significantly increased,JAK2 the phosphorylation of STAT3 was significantly enhanced.Conclusions:(1)6 weeks aerobic exercise can promote the expression of MyoD and Myf5 in skeletal muscle,and can not activate the JAK2/STAT3 signaling pathway.(2)6 weeks resistance exercise rats significantly increased body weight,lean body weight increase,and promote the expression of skeletal muscle related MyoD,Myf5 development through JAK2/STAT3 signaling pathway,and affect the growth and development of skeletal muscle.(3)The effect of 6 weeks resistance exercise in promoting MyoD,Myf5 gene expression is better than aerobic exercise for 6 weeks.
