Objective: To study the effect of moderate intensity aerobic exercise on the regulation of STAT3 / SOCS3 signal pathway by mir-125a-p in the liver of mice with obstructive jaundice.Methods: Methods: 30 healthy male KM mice were randomly divided into three groups: sham operation(OS)group,obstructive jaundice model(OM)group and obstructive jaundice + aerobic exercise(OT)group.In OM group and ot group,the right angle suspension of common bile duct was used to construct the animal model of obstructive jaundice.In OT group,0% gradient and 10 m / min no load training(30 min / day)were carried out,and the running plan of 6 weeks was maintained.After six weeks of intervention,the serum and tissue of mice were collected,the liver tissue was sliced,the morphological changes of liver cells were observed,he staining was used;the levels of TBIL,alt,AST,TC and TG were detected by blood biochemical liver function;STAT3 was determined by immunohistochemistry/ The protein levels of STAT3,SOCS3 and SREBP-1c,which are related to lipid metabolism downstream of SOCS3 channel,were differentially expressed in the liver tissues of mice.The mRNA levels of mir-125a-5p,STAT3 and SOCS3,SREBP-1c and FASN mRNA related to lipid metabolism were detected by realtime PCR.Results:(1)HE staining showed that the morphology and structure of hepatocytes were normal in OS group,extensive vacuolar degeneration and lamellar necrosis of hepatocytes were found in OM group,the structure of hepatic lobules was basically normal in OT group,and the arrangement of hepatocyte cords was relatively neat.(2)Serological results: the levels of TBIL,ALT and AST in the OT group were lower than those in the OM group;the levels of TG,TC and LDL in the OM group were lower than those in the OS group,and the levels of TG,TC and LDL in the OT group were lower than those in the om group(P < 0.05),and the levels of HDL in the OT group were higher(P < 0.01).(3)The results of immunohistochemistry showed that the expression of STAT3 and SOCS3 in liver tissue of OT group was significantly lower than that of OM group(P < 0.01),and the expression of srebf-1c in liver tissue of OT group was significantly lower than that of OM group(P < 0.01).(4)QRT-PCR results showed that the expression level of mir-125a-5p in OM group was significantly lower than that in OS group(P < 0.01),and the expression level of mir-125a-5p in OT group was significantly higher than that in OM group(P < 0.01),indicating that mir-125a-5p may participate in the regulation of liver lipid metabolism in OJ mice,and aerobic exercise may affect the expression level of mir-125a-5p;the expression of STAT3 and SOCS3 in OM group was significantly higher than that in OS group(P< The expression of STAT3 and SOCS3 in OT group was significantly lower than that in OM group(P < 0.01);the expression of SREBP-1c and FASN in OM group was higher than that in OS group(P < 0.01),and the expression of SREBP-1c and FASN in OT group was lower than that in OM group(P < 0.01).Conclusion: Moderate-intensity aerobic exercise has a better effect on improving lipid metabolism disorders in mice with OJ liver injury;Affect the expression of downstream target genes,and ultimately improve OJ liver injury-induced lipid metabolism disorders.
