Objective: The purposes of this study were to explore the effects of exercise intervention(4 weeks)of growth and development period and long-term aerobic exercise intervention(12 weeks)on FTO and related gene expression in adipose tissue of rats and to explore the molecular mechanism of exercise regulation of fat metabolism.Methods: Forty three-week-old SD male rats were randomly divided into control group(C4,C12)and exercise group(E4,E12),and there were 10 rats in each group E4?E12 group were participated in treadmill training at 60% VO2 max for4 weeks?12 weeks,and the speed of treadmill based on the determination of the maximum oxygen uptake.The training were carried out 5 days a week,1 hour per day.The body weight and food intake were recorded weekly.The blood lipid ?perirenal and epididymal adipose tissue weights of the rats in the fasting state after training were measured.The expressions of FTO and C/EBP?,PPAR?,FAS,Perilipin,HSL mRNA in adipose tissue and lipid metabolism-related gene were measured by RT-PCR.The protein content of FTO,C/EBP? and PPAR? in adipose tissue was determined by western blot.Results:1,With the increasing of age,the 4 groups rats body weight were growing,and the E4 group was significantly lower than that of C4 group at the corresponding time points from the second week of exercise until the end of 4th week exercise;The body weight in E12 group was significantly lower than that of C12 group until the end of the 12 th weekexercise;2?The visceral fat absolute weight and relative weight of rats in E4,E12 group were significantly lower than that C4,C12 group;3?The levels of serum LDL-C in group E4 was significantly lower than that in group C4;HDL-C was significantly higher in group E12 than that in group C12,and serum HDL-C was significantly higher than that in group C12;4?The protein expression of FTO and PPAR? in E4 group was significantly lower than that in C4 group,and the expression of C/EBP?,PPAR?,FAS and Perlipin mRNA in E4 group was significantly lower than that in C4 group,and the expression of HSL mRNA in E4 group was significantly higher than in C4 group;The expression of FTO,C/EBP?,PPAR?,FAS and Perilipin mRNA in adipose tissue of rats were significantly lower than that in C12 group.Conclusion : 1.Aerobic exercise intervention(4 weeks)of growth and development period inhibit adipocyte differentiation and fat synthesis by down-regulating the expression of FTO,PPAR?,FAS gene in adipose tissue;playing a role by promote lipolytic gene Perilipin and HSL,and reducing the body weight and lipid ratio 2.12 weeks aerobic exercise at childhood can inhibit fat synthesis,decrease body weight and body fat ratio by down-regulating FTO,C / EBP?,PPAR?,FAS mRNA levels...
