Skeletal muscle is one of the biggest tissues and organs of human,and is the power of the body for all kinds sports.At the same time,is also the biggest metabolic and endocrine organs.Before,a large number of studies have shown that the energy metabolism of skeletal muscle is closely related to a variety of met abolic syndrome,what`s more,the muscle metabolic disorders can lead to the body be in a variety of pathological conditions,such as obesity,type 2 diabetes,myophagism,myasthenia gravis and so on.there is no doubt that it is brings serious harm to the health of the body.Skeletal muscle contraction can also through some form,such as endocrine,and autocrine,secrete the muscles of the factors with bioactive,such a IL-15.And also,these muscles factors can regulate the energy metabolism of skeletal muscle.In addition,the motion of skeletal muscle contraction,both aerobic and an aerobic conditions will produce lactic acid,lactic acid is the inevitable outcome of the skeletal muscle contractions,can measure the contraction of skeletal muscles state.Therefore,lactic acid stimulation can be used as a simulation movement conditions of one of the classic indicators.But,the content of lactic acid and the concentration have a great difference,because of different exercise intensity and exercise load.So,it showed that different lactic acid concentration and stimulation time may lead to different effects,that also namely to muscle factor IL-15 different adjustment.In order to research the condition of lactic acid stimulation to simulate movement at different time on muscle lactic acid concentration and stimulating factor IL-15 m RNA expression level in skeletal muscle.In this study,we designed the concentration of lactic acid with 8m M and 16 m M and different stimulus time of sodium lactate with 1 hour and 4 hours,to evaluate the effects of the different concentration and time on muscle factor IL-15 m RNA.Objective: Built the model of lactic acid stimulation simulated in movement,evaluated the expression level of sport factor IL-15 m RNA in different lactic acid concentration and stimulation time.Method: Cultivation C2C12 cells of mice in vitro,induced differentiation of C2C12 cells in mice to the normal muscle differentiation,to be six days until cell differentiation appeared muscle tube,with a tendency for 8 and 16,respectively,tendency for concentration of lactate processing cell 1 hours and four hours,and to set up blank control group.Learn from lactate treated cells for real-time PCR,test the different influence of IL-15 m RNA expression level in C2C12 cells with different concentration.Results:(1)After lactic acid stimulation 1h,the IL-15 m RNA expression of 8m M and 16 m M concentration group were slightly lower than control group(27.64±0.81,27.68±0.14 vs.28.21 ±0.25,P>0.05),no statistical difference;Compared to the 8m M and16 m M concentration group(27.64±0.81 vs.27.68±0.14P>0.05),the expression of IL-15 of 16 m M concentration group was slightly higher than the 8m M concentration group,but no statistical difference;(2)After lactic acid stimulation 4h,the IL-15 m RNA expression of 8m M and 16 m M concentration group were slightly higher than control group(31.32±0.33,34.47±0.42 vs.28.21±0.25,P<0.01),a very significant difference;Compared to the 8m M and16 m M concentration group(31.32±0.33 vs.34.47±0.42P<0.01),a very significant difference.Conclusion:(1)we were built the simulated model of different intensity and time of movement by the lactic acid simulation;(2)After 1h lactic acid stimulation,the IL-15 m RNA expression of C2C12 cells were not obvious;after 4h lactic acid stimulation has a promoting effect on its expression,and the 16 m M concentration was more effective than the 8m M concentration;(3)This study reminded,exercise induced factor IL-15 m RNA expression is associated with the intensity of movement,more obvious under big strength;(4)This study reminded,exercise induced factor IL-15 m RNA expression is associated with the duration of movement,more obvious under long time.
