Objective: The effect of 4 weeks of low intensity treadmill exercise on the brown adipose tissue(BAT)of obese mice would be observed in this study.The key differentially expressed genes related to glucose-lipid metabolism screened by gene expression profile were verified by RT-q PCR,in order to explore the mechanism of aerobic exercise effect on glucose-lipid metabolism in BAT of obese mice.Methods: 30 weaning C57BL/6J male mice were randomly divided into the normal diet control group(Group N,n=8),fed with the maintain fodder,and the high-fat diet group(Group HD,n=22),fed with high-fat fodder with 40% fat.The model of obesity was established after 8 weeks,the group HD were randomly subdivided into 2 groups: the obese control group(Group OB,n=8)and the obese exercise group(Group E,n=8).The group E was trained at a speed of 10m/mim,1 h/day and 6 times/wks,for 4 weeks.Their body weights and food intakes were recorded every week.After 4-week intervention,serum lipid(TG and CHO)and glucose(GLU)were detected;Microarray hybridization of mRNA from BAT of scapula was scanned to generate gene lists with corresponding fluorescence intensity data.Genes were significantly(p?0.05)up-or down-regulated more than 2 fold were chosen to do Gene Ontology and enrichment analysis by KOBAS2.0.The genes,Acsl5,Fasn,Acaca,Fabp5,Ptges,Ptgis and Fbp1,participating in key biological process(BP)or PPAR,AMPK,Insulin and VEGF pathway were confirmed by RT-q PCR.Results:(1)Comparing with group N,there are 445 genes were significantly up regulated and 796 were down regulated.The BP of these genes are mainly about the synthesis and metabolism process of lipids,sterol,cholesterol and steroid,as well as the chemotaxis of hemameba and granulocyte.Comparing with group OB,there are 486 genes were significantly up regulated and 286 were down regulated.The BP of these genes are mainly about the synthesis and metabolism process of lipids,sterol,cholesterol and steroid,as well as the metabolism process of carboxylic acid,acetone acid and coenzyme,and antioxidant process.(2)The pathways that aerobic exercise regulate the function of BAT were mainly involving PPAR,AMPK,Insulin and VEGF signaling pathway.(3)Choosing Acsl5,Fasn,Acaca,Fabp5,Ptges,Ptgis and Fbp1 as the important genes.(4)The levels of Acsl5,Fasn,Acaca,Fabp5 and Ptgis mRNA in group E were significantly higher than group OB,Ptges and Fbp1 mRNA levels were slightly higher than OB.Conclusion: The process of glucose and lipid metabolism in BAT of obese mice probably be regulated by PPAR,AMPK,insulin and VEGF signaling pathway by aerobic exercise,to help improve the BAT activity,and effectively reduce the obese mice body weight and blood glucose and lipid levels,and finally promote the body's health.
