Objective:In this study,obesity was induced by high-fat diet in mice,and the expression of Sirt1 in exercise and dietary intervention in high-fat diet mice and its role in promoting lipid hydrolysis were observed,so as to provide theoretical basis for the transformation of lipid metabolism disorder and the prevention and treatment of obesity.Methods:Male C57BL/6 mice of 4 weeks old,a total of 50 rats,mice were reared in the standard experimental animal room for 3 days.Due to the death,they were randomly divided into the normal control group(NC,N=8)to feed the basic feed.The obese group was fed high-fat diet for modeling for 10 weeks.After successful modeling,they were randomly divided into the obese group(HC,N=8),who did not participate in sports training and continued to feed high-fat diet.In the obese diet control group(HN,N=8),was fed the 70% high-fat control diet,and no exercise training was involved.In the obesity exercise group(HE,N=8),aerobic exercise intervention was performed and high-fat diet was fed.In the obese diet control exercise group(HNE,N=8),aerobic exercise intervention was performed,and the 70% highfat control group diet was fed.After sampling,Serum FFA was determined by ELISA,protein expressions of Sirt1,PPARγ,Perilipin and HSL in adipose tissue were detected by western-blot method,and m RNA expressions of Sirt1,PPARγ,Perilipin and HSL in adipose tissue were determined by real-time quantitative fluorescence PCR.Results:1.Compared with NC group,body weight,abdominal fat weight and lipid body ratio were significantly increased in HC group(P<0.01).Compared with HC group,the body weight,fat weight and lipid body ratio of HN group and HNE group decreased significantly(P<0.01),while the body weight and fat content of HE group decreased significantly(P<0.01);Compared with the HN group and HE group,all the three indexes of HNE group were significantly decreased(P<0.01).2.Compared with NC group,serum FFA in HC group was significantly increased(P<0.01).Compared with HC group,serum FFA in HE,HN and HNE group decreased significantly(P<0.05,P<0.05,P<0.01).3.Compared with the NC group,the protein expression of Sirt1 and HSL in the adipose tissue of the HC group decreased significantly(P<0.01,P<0.05),while the protein expression of PPARγ and Perilipin increased significantly(all P<0.01).Compared with HC group,the protein expression of Sirt1 and HSL in adipose tissue of HE group,HN group and HNE group were significantly decreased and increased(P<0.05 or P<0.01),the protein expression of PPARγ was significantly decreased(P<0.05 or P<0.01),and the protein expression of Perilipin in HE group and HNE group was significantly decreased(P<0.05 or P<0.01),Compared with HN in HE group,PPARγ protein expression in adipose tissue of mice in HNE group decreased significantly(P<0.05 or P<0.01).4.Compared with NC group,the relative expressions of Sirt1 and HSL m RNA in the adipose tissue of HC group were significantly decreased(P<0.01,P<0.05),while the expression levels of PPARγ and Perilipin m RNA increased significantly(P<0.01).Compared with the HC group,the relative expressions of Sirt1 and HSL m RNA in the adipose tissue of the HE and HNE groups were significantly increased(P<0.01 or P<0.05),and the relative expressions of PPAR γ and Perilipin m RNA were significantly decreased(P<0.01 or P<0.05).The relative expressions of Sirt1 and HSL m RNA in the HN group were significantly increased(P<0.01 or P<0.05),and the relative expressions of PPARγm RNA was significantly decreased(P<0.01);Compared with the HE group,the relative expression of Sirt1 and HSL m RNA in the HNE group was significantly increased(P<0.05,P<0.01),and PPARγ lifting m RNA was significantly decreased(P<0.01).Compared with the HN group,the relative expression of Sirt1 and HSL m RNA in the HNE group was significantly increased(P<0.05,P<0.01),and the expression of PPARγ pulling and Perilipin m RNA was significantly decreased(P<0.05,P<0.01).Conclusion:1.Long-term high-fat diet can cause obesity in mice,decrease Sirt1 expression in adipose tissue,weaken the control effect of PPAR γ /Perilipin,increase Perilipin,protect triglycerides from being broken down by HSL,cause lipid deposition,and cause fat metabolism disorder.2.After 8 weeks of exercise and dietary intervention,the expression of Sirt1 in the adipose tissue of mice could be increased and then the PPARγ /Perilipin signaling pathway was down-regulated.The down-regulation of Perilipin weakened the protective role of the barrier against fat,and the up-regulated expression of HSL promoted the strengthening of fat decomposition,reduce fat deposition and transform lipid metabolism disorder.Compared with the dietary intervention,the effect of exercise on Perilipin is more obvious and the effect of fat catabolism is more significant.
