We research the effects and mechanisms of caffeine to promote the sports capacity from sports practice and cell biology based on nuclear magnetic resonance(NMR)technology:1)using cell metabonomics,analysis the effects of caffeine on C2C12 myoblasts and myotubes metabolic patterns to explain the action mechanism of caffeine on skeletal muscle;2)using urine metabonomics,analysis the difference of urine metabolic pattern of basketball players with/without caffeine to explain the mechanism of caffeine improving sports capacity.The introduction mainly summarizes the research questions,and carries on the elaboration to the research mentality of this article.The first chapter we studied the effects of caffeine on C2C12 myoblasts and myotubes metabolism under different nutritional conditions.Principal component analysis(PCA)results show:in nutrient rich conditions,caffeine has no effect on the metabolism of skeletal muscle cells;under the condition of insufficient nutrition,caffeine induced metabolism of skeletal muscle cells changed obviously and make the metabolic pattern close to normal group.Metabolites and metabolic pathway analyses revealed the molecular mechanism of caffeine on C2C12 myoblasts and myotube,namely caffeine induced changes in energy metabolism,glycerophospholipid metabolism,glutathione biosynthesis and TCA.Meanwhile,the results showed that the mechanism of caffeine on C2C12 myoblasts and myotube are not exactly the same.In the second chapter,we studied the effects of supplementation/not supplementation of caffeine on the urine metabolic pattern of basketball players.The results showed that the anaerobic glycolysis was the main energy supply function of the high-intensity intermittent training.After the training,the concentration of lactic acid,butyric acid,hydroxy butyric acid and pyruvic acid in urine changed obviously,which could be used as a biomarker for the training of mixed oxygen ability.Caffeine increased the fatty acid metabolites in urine and decreased the concentration of glycine.Metabolic pathway analyses showed that caffeine supplement reduces the metabolic pathways associated with glucose metabolism,strengthen TCA metabolism,pyruvate metabolism and mitochondrial electron transport,improved the level of aerobic metabolism capacity and the overall energy metabolism.The third chapter is a case study.We choose a elite player who are sensitive to caffeine to study the difference of urine metabolic pattern with/without caffeine.Caffeine reduce the difference of before and after the training to achieve the energy of saving and maintain the homeostasis of body metabolism during exercise.
