Objective:In this study,in the 2015 Taiyuan International Marathon amateur as the research object,the application of 1H NMR(nuclear magnetic resonance)group of metabolic marathon amateur urine metabolomics method combined with urine routine long-term endurance training monitoring system,to make clear training in vivo metabolites,metabolic compounds are closely related to the marathon,from the level of long-term endurance training for metabolite monitoring to provide new ideas.Methods:We selected to participate in the 2015 Taiyuan International Marathon 20 amateur male players,players between the ages of 41-45,in the morning urine collection of marathon amateur players to participate in the 2015 Taiyuan international marathon match quiet(urine),urine collection 20-50ml again in the players after the game,this paper uses literature method,experimental method and mathematical statistics.And the logical analysis method,first routine urine test on the urine,and then use 1H NMR(nuclear magnetic resonance)metabonomics method for metabolomics experiments,based on the MestReNova-6.1.1 software,the data is first baseline and phase adjustment,and multivariate statistical analysis using SMICA-P13.0(Umetric,Sweden)were treated by partial least squares projection discriminant analysis(PLS-DA)by analyzing the differences of each group.Further single factor variance analysis was performed using SPSS 17.0 on different metabolites.Results:(1)According to the urine routine test before and after the experiment,the results showed that the urine routine test before and after the match was different,especially the detection rate of urinary protein and the detection rate of urinary bladder had significant changes.In the marathon,the positive rate of PRO at rest and after the game had significant difference(P<0.05),the positive rate of URO at rest and after the game had significant difference(P<0.05).In the half marathon,the positive rate of PRO at rest and after the game had significant difference(P<0.05).The highest positive rate of GLU in the quiet,after the highest detection rate of BLD positive.The full and half marathon,PRO positive detection rate is the highest at the end of the game,URO positive detection rate is the highest at the end of the game,BIL positive detection rate is the highest in the game.(2)According to the detection of urinary metabolites before and after the experiment the body of the game,marathon amateurs,10 with significant difference(P<0.05)of potential biomarkers,and compared the game before,alanine,acetamide,glycoprotein two lipid,methylamine,lysine,glycine,citrulline content decreased,while benzene acetylglycine,glycerin,inositol content increased(P<0.05,P<0.01).Half marathon amateurs,before the game fell in the same area of urinary metabolites in Score Plot,including the scattered points in information does not exist differences.Conclusion:(1)Half marathon amateurs before and after games in urine protein index were significantly different,metabolomics results showed no significant differences in metabolite after the game before the half marathon,not statistically significance.(2)There were significant differences in the full marathon amateurs urine protein and urobilinogen indicators before and after the game in the urine,metabolomics metabolites display in addition to protein,there is a big difference metabolites involved in sugar,fat and amino acid,propionic acid,acetamide,glycoproteins,lipids,dimethylamine,lysine,citrulline,glycine phenylacetyl,inositol,glycerol and glycine metabolism pathway may be potential biomarkers.Metabolite before the full marathon after the game there was a significant difference,indicating that the marathon is aerobic energy supply-based movement,and not only the aerobic oxidation of carbohydrate,protein and fat and aerobic oxidation can also occupy a certain percentage.(3)Marathon amateur marathon should pay attention not only to timely nutritional supplement sugar depending on the distance of the race,but also the nutritional supplement of protein and fat.
