Objective: To study effects of hypoxia and leucine intervention on the activation of myocardial Sestrin2/AMPK? 2 signaling pathway in exercise rat,and possible mechanism under this procession.Methods: Experiment 1:rats were subjected to either a single bout of exercise or four weeks exercise training and then euthanized at 0h,3h,24 h post exercise.experiment 2:rats were subjected to different exercise load,and exposed to either normoxia or hypoxia,and then euthanized immediately post exercise.experiment 3: rats were subjected to either leucine supplement with a single bout of exercise or four weeks exercise training with leucine supplement,and then euthanized either immediately post exercise or 24 h post exercise.Results:(1)experiment 1: Sestrin2 in left ventricular muscle protein expression and AMPK? 2 activation levels were significantly increased immediately after exercise.There was no significant change in the expression of Sestrin2 protein and the activation of AMPK? 2 in the left ventricle after the exercise in the four-week trained rats.(2)experiment 2: the expression of Sestrin2 and the activation of AMPK? 2 in the left ventricle were significantly increased in exhausted exercised rats.(3)experiment 3: in the supplemental leucine acute exercise model,the expression of Sestrin2 and the activation of AMPK? 2 in the left ventricle were significantly increased after acute exercise.However.The expression of Sestrin2 and the activation of AMPK? 2 in the left ventricle were no significant change after leucine supplement with acute exercise.Conclusion:(1)acute exercise transiently increased the activation of Sestrin2/AMPK? 2 signaling pathway,long-term endurance training has no effect on the resting level of the Sestrin2/AMPK? 2 signaling pathway.(2)the Sestrin2/AMPK? 2 signaling pathway was closely related to exercise stress,while hypoxic intervention had no obvious effect on the activation of the myocardial Sestrin2/AMPK? 2 signaling pathway.(3)leucine supplementation can inhibited the stimulation of acute exercise on the myocardial Sestrin2/AMPK? 2 signaling pathway.No significant change of the Sestrin2/AMPK? 2 signaling pathway activation were observed in the rat myocardium after four weeks training and leucine supplementation.
