Objective:To investigate the effects of Bushen Huoxue prescription and transplanted microencasulated granulosa cells on rats reproductive and endocrine systems.Methods:Total 60 SD female rats were randomly divided into six groups:the blank group?A group?,the model group?B group?,microencapsulated granulosa cells transplanted group?C group?,the low dose Bushen Huoxue prescription and microencapsulated granulosa cells transplanted group? D group?,the middle dose Bushen Huoxue prescription and microencapsulated granulosa cells transplanted group?E group?,the high dose Bushen Huoxue prescription and microencapsulated granulosa cells transplanted group? F group?.After 28 days treatment,the rats were sacrificed and taken material.Enzyme-linked immune sorbent assay?ELISA? was adopted to detect the concentrations of serum estradiol?E2?,follicle stimulating hormone?FSH?,and luteinizing hormone?LH?.Results: B group weights and ovarian index less than other five groups, significant diffenrence?P<0.05?,F group ovarian index is higher than B group, significant diffenrence?P<0.05?.In D group,E group and F group weight increases,but the difference is not statistically significant?P>0.05?.After removal of the ovaries, B group E2 level less than A group obviously?P<0.01?, whereas FSH and LH levels increased significantly?P<0.05?.Compare C group with B group, FSH and LH levels decreased, E2 level increased, significant diffenrence?P<0.05?;FSH and E2 level between A group and F group have no statistically difference?P>0.05?.FSH,LH,and E2 level has difference between C group and F group?P<0.05?.Conclusion:Compared with simple transplantation of microencapsulated granulosa cell,Bushen Huoxue prescription and transplantation of microencapsulated granulosa cell in ovariectomized rats can better improve hormone secretion condition,provide a good prospect for Chinese medicine of clinical prevention and treatment of premature ovarian failure. |
