Objective:To explore the effect of Bushen Huoxue Prescription on the expression of PI3K,AKT1 and MTOR in mice with immune premature ovarian failure?POF?.Methods:SPF Balb/c female mice were injected subcutaneously with zona pellucida 3 as antigen to establish an immune premature ovarian failure model.After successful preparation,the mice were randomly divided into model group,positive group,high,medium and low concentration groups of Bushen Huoxue recipe.The drug was administered continuously for 30 days.The histopathological changes of ovary were observed by hematoxylin-eosin?HE?staining under light microscope.The levels of estradiol?E2?,follicle stimulating hormone?FSH?and luteinizing hormone?LH?were detected by ELISA.The expression of PI3K,AKT1 and MTOR protein in ovary was detected by immunohistochemistry and analyzed by statistics.Result:?1?Compared with the blank group,the body weight of the model group was significantly lower?P<0.01?.After HE staining of vaginal exfoliated cells,estrous cycle disorder was found in the model group,suggesting that the model was successful.The hormone level was detected by ELISA,which provided evidence for the success of the model.?2?Compared with the blank group,the levels of E2 and FSH and LH in the model group decreased,while the levels of E2 and FSH and LH increased?P<0.01?.Compared with the model group,the levels of E2 and FSH and LH in both the Chinese herbal medicine group and the positive group increased,and the effect of the high concentration group of Chinese herbal medicine was the best?P<0.01?.?3?Compared with the blank group,the index of ovary and thymus in the model group decreased significantly?P<0.01?,while the index of spleen increased significantly?P<0.01?.Compared with model group,ovary and thymus index of positive group and high concentration group of traditional Chinese medicine increased significantly?P<0.01?,while spleen index decreased?P<0.01?.There was no significant difference in ovary and thymus index between the middle concentration group and the low concentration group?P>0.05?,but there was significant difference in spleen index?P<0.05?.?4?Compared with the blank group,the expression of PI3K,AKT1 and MTOR in ovarian tissue of the model group decreased significantly?P<0.01?.Compared with the model group,the number of mature follicles and atresia follicles in each concentration group and positive group increased significantly,and the expression of PI3K,AKT1 and MTOR protein in high concentration group and positive group increased significantly?P<0.01?.Conclusion:Bushen Huoxue Prescription may improve the ovarian function of POF mice by affecting the expression of PI3K,AKT1 and MTOR proteins,promote the growth and development of follicles,increase the expression level of E2,reduce the expression level of FSH and LH,and maintain the stability of the ovarian internal environment.
